This study focused on an 80% ethanol:water extract of Galenia africana and Dicerothamnus rhinocerotis in which a phytochemical study revealed the presence of flavonoids as the major secondary plant metabolites. Eleven pure flavonoids viz., (E)-2',4'-dihydroxychalcone 1, (S)-7-hydroxyflavanone 2, (E)-2',4'-dihydroxy-2,3-dihydrochalcone 3, (S)-5,7-dihydroxyflavanone 4, (S)-2',5,7,-trihydroxyflavanone 5, (S)-5,7-dihydroxy-2'-methoxyflavanone 6, 5,7-dihydroxy-4H-chromen-4-one 7, (S)-5-hydroxy-7-methoxyflavanone 8 and (E)-2-hydroxy-3',6'-dimethoxychalcone 9 were isolated from G. africana, while [sakuranetin] (S)-4',5-dihydroxy-7-methoxyflavanone 10 and [eriodictyol-3',7-dimethyl ether] (S)-4',5-dihydroxy-3',7-dimethoxyflavanone 11 were isolated from D. rhinocerotis. Compounds 6 and 9 are new while this is the first reported isolation of 1, 2, 3, 4, 5, 7, 8, 10 and 11 from these plants. All isolated compounds were tested for their antimycobacterial activity against the reference strain Mtb H37Rv. The most active compound, 9, demonstrated a MIC 99 of 5 µM against Mtb H37Rv American Type Culture (ATCC) and (ATCC27294), which were also sensitive to Isoniazid (INH) and Rifampicin. The antibacterial activity of 9 might be ascribed to the presence of features such as the α,β-unsaturated ketone and the substitution patterns on the A and B rings.

The indiscriminate use of non-regulated skin lighteners among African populations has raised health concerns due to the negative effects associated with skin lightener toxicity. For this reason, there is a growing interest in the cosmetic development of plants and their metabolites as alternatives to available chemical-derived skin lightening formulations. Approximately 90% of Africa’s population depends on traditional medicine, and the continent’s biodiversity holds plant material with various biological activities, thus attracting considerable research interest. This study aimed to review existing evidence and document indigenous African plant species capable of inhibiting the enzyme tyrosinase and melanogenesis for potential incorporation into skin lightening products. Literature search on melanin biosynthesis, skin lightening, and tyrosinase inhibitors resulted in the identification of 35 plant species were distributed among 31 genera and 21 families across 15 African countries and 9 South African provinces. All plants identified in this study showed competent tyrosinase and melanogenesis inhibitory capabilities. These results indicate that African plants have the potential to serve as alternatives to current chemically-derived skin lighteners.

The reproductive health of individuals is severely compromised by HIV infection, with candidiasis being the most prevalent oral complication in patients. Although not usually associated with severe morbidity, oropharyngeal candidiasis can be clinically significant, as it can interfere with the administration of medications and adequate nutritional intake, and may spread to the esophagus. Azole antifungal agents are commonly prescribed for the treatment and prophylaxis of candidal infections, however, the emergence of drug resistant strains and dose limiting toxic effects has complicated the treatment of candidiasis. Consequently, safe and effective and affordable medicine is required to combat this fungus. Commercial garlic (Allium sativum) has been used since time immemorial as a natural antibiotic, however, very little is known about the antifungal properties of two indigenous South African species of garlic, namely Tulbaghia alliacea and Tulbaghia violacea, used as folk medicines for a variety of infections. This study compares the in vitro anticandidal activity of Tulbaghia alliacea, Tulbaghia violacea and Allium sativum extracts. It was found that the greatest concentrations of inhibitory components were extracted by chloroform or water. The IC50 concentrations of Tulbaghia alliacea were 0.007-0.038% (w/v). Assays using S. cerevisiae revealed that the T. alliacea extract was fungicidal, with a killing half-life of approximately 2 h. This inhibitory effect of the T. alliacea extracts was observed via TLC, and may be due to an active compound called marasmicin, that was identified using NMR. This investigation confirms that extracts of T. alliacea exhibit anti-infective activity against candida species in vitro.
