Tying up loose ends: Oligoamide thread molecules can spontaneously transform into open knots, whose terminal functional groups can subsequently be chemically linked to form closed, cyclic knots (see scheme; left: threads; middle: open knots; right: molecular knots; X=N, C).
New topological chiral molecular knots (knotanes) 3a–6 of the amide type were substituted with long aliphatic side chains (butyloxy, hexyloxy, octenyloxy, decyloxy) and with p‐bromobenzyloxy and oligoethylene glycol ether units, which resulted in improved solubility and enhanced potential for racemate separation. In consequence, the knotanes with longer substituted chains could be separated even on commercially available noncovalently bound chiral stationary phases. The (p‐bromobenzyloxy)‐knotane 6 and the octenyloxy‐knotane 4 were synthesized with the intention of using their functional sidechains in further reactions, 6 being used in Suzuki‐type reactions, while 4 was consequently allowed to react in the presence of Grubbs catalyst in metathesis reactions. The achieved solubility allowed us to test the abilities of the alkyl chain‐substituted knotanes 3a–c and the (p‐bromobenzyloxy)‐knotane 6 to form Langmuir–Blodgett films. The assemblies were investigated on mica by AFM technique. A key point of focus was the (topological) chirality of the long chain‐substituted knotanes. To address this point, the racemates were enantioseparated by HPLC on various chiral phases, and the obtained pure enantiomers were found to exhibit pronounced Cotton effects in their circular dichroism spectra. Professor Vincenzo Balzani zum 70. Geburtstag gewidmet Oligoamid-basierte molekulare Knoten waren bisher nur durch intermolekulare Eintopfkondensation von drei Diamin-(1) und drei Säurechloridmolekülen (2) zugäng-lich [1][2][3] (Schema 1, Route A). Als Mechanismus der Verknotung hatten wir die intermediäre Bildung von längeren Oligoamidfäden 3 a oder 4 angenommen.[2] Aus Röntgenstruk-turanalysen von Amidknoten [2] des Typs 6 und weiteren experimentellen und theoretischen Daten [4] schlossen wir, dass das Falten von linearen Fadenvorstufen wie 3 a oder 4 (Schema 1, Routen B, C) zu verknoteten Fadenfragmenten dank günstiger Wasserstoffbrücken-Bindungsmuster in nichtkompetitiven Lösungsmitteln (z. B. Dichlormethan) bereits programmiert sein könnte.[5] Die intermediäre Bildung von 3 b aus 5 können wir allerdings nicht ausschließen, wenn auch die Reaktionsbedingungen (Stöchiometrie der Zugabe von 2 a: 1 Moläquiv. bei Route B, 2 Moläquiv. bei Route C) diese Annahme nicht stützen. Wir berichten hier über die erstmalige Synthese, Isolierung und Charakterisierung der Fäden 3 a und 4 sowie ihre Umsetzung zu den Knotanen 6 (Schema 1).Die Synthese der verlängerten Fäden 3 a und 4 als mög-liche Vorstufen der Amidknotenbildung ist in Schema 2 skizziert. Damit ergibt sich die Möglichkeit, zwischen den Routen B und C zu unterscheiden, indem die isolierten Fäden 3 a bzw. 4 getrennt mit 2 bzw. mit 1 und 2 umgesetzt werden und anschließend geprüft wird, ob ein molekularer Knoten 6 entsteht.Route B: Der Faden 3 a faltet sich von selbst, anschließend fädelt er sich intramolekular durch die gebildete Schleife und bildet spontan den (einfach) offenen Knoten 3 b[6] (Schema 1, Route B). Um diesen Vorgang nachzuweisen, haben wir das isolierte Decaamid mit 4-substituierten Pyridindicarbonsäuredichloriden (2 a-d) umgesetzt (Schema 3), wobei der unverknotete Faden 3 a einen achiralen Makromonocyclus 7 und der offene Knoten 3 b einen isomeren, geschlossenen, topologisch chiralen Knoten 6 mit drei Pyridineinheiten liefern sollte. Diese Reaktion bietet zugleich die Möglichkeit, durch Einsatz eines substituierten Pyridindicarbonsäuredichlorids 2 im letzten Schritt erstmals monosubstituierte Knoten 6 a-d herzustellen (Schema 3).Einerseits hilft diese Strategie also, den Templatmechanismus der Verknotung von neutralen (ungeladenen) Molekülen (ohne Hilfe von Kationen [3] ) zu verstehen, und andererseits lassen sich mit ihr neue Kleeblattknoten mit unterschiedlichen Bauelementen synthetisieren. Eine derartige spontane Selbstverknotung 3 a!3 b von niedermolekularen synthetischen Fadenmolekülen im präparativen Maßstab ist bisher unseres Wissens noch nicht beschrieben worden. [7] Die Umsetzung des langen Fadens 3 b mit unsubstituiertem Pyridindicarbonsäuredichlorid (2 a) führt in der Tat zum bereits früher von uns nach Route A synthetisierten unsubstituierten Knoten 6 a mit drei gleichartigen Pyridineinheiten, was in Einklang mit dem von uns vorgeschlagenen Mechanismus der Route B ist.
