A longitudinal study of the infection dynamics of Salmonella enterica was carried out with three Danish farrow-to-finish swine herds. To account for variations in Salmonella shedding over time, litters from each herd were divided into two cohorts. Each cohort consisted of 30 pigs, for a total of 180 pigs. Pigs were individually monitored by monthly bacteriologic and serologic examinations from weaning to slaughter. At weaning, individual sows were examined bacteriologically and serologically. At slaughter, cecal contents, ileocecal lymph nodes, and carcass swab samples were obtained from 131 pigs. A total of 88 pigs were found to be shedding Salmonella on one or more occasions. Only the Salmonella serotype Typhimurium was detected during the study period. At weaning, no sows or piglets were found to be shedding, but a serological reaction was detected in 11 sows. The prevalence in culture peaked in the nursery and subsequently declined to undetectable levels before slaughter. The seroprevalence peaked approximately 60 days after the peak prevalence in culture. Salmonella was detected in individual fecal samples at least once in 53% of the pigs, and 62% of the pigs were seropositive more than once. Only 3.7% of all pigs were found to be culture positive on more than one occasion. Piglets from seroreacting sows had a significantly (P ‫؍‬ 0.0339) lower probability of shedding in the nursery. Under the assumption that shedding lasted at least 1 or 2 weeks, the average shedding time was estimated to have been 18 or 26 days. An association between serology, on-farm bacteriology, and Salmonella prevalence in culture at slaughter was shown. Marked differences in prevalence in sera and prevalence in culture between cohorts and within herds were observed. These differences emphasize the need for caution when using point estimates in on-farm interventions and surveillance in subclinically infected swine herds.Salmonella enterica does not normally cause clinical disease in pigs, but subclinical Salmonella infections constitute an important food safety problem throughout the world. From a consumer viewpoint, continuing efforts are needed to reduce the occurrence of Salmonella in pork. In order to achieve this, information about the dynamics of Salmonella infections in swine herds over time (e.g., duration of infection and disease transmission patterns) can be a useful tool. A preliminary epidemiological survey indicated an association between Ascaris infections in Danish patients and contact with pigs or pig manure. In the present study, we compared Ascaris worms collected from humans and Ascaris worms collected from pigs by amplified fragment length polymorphism (AFLP) analysis, a technique for whole-genome fingerprinting, and by PCR-linked restricted fragment length polymorphism (PCR-RFLP) analysis of the internal transcribed spacer region of nuclear rDNA. The AFLP data were analyzed by distance-and model-based clustering methods. These results assigned Ascaris worms from Danish patients to a cluster different from that for worms from humans in other geographic areas. In contrast, worms from humans and pigs in Denmark were assigned to the same cluster. These results were supported by the PCR-RFLP results. Thus, all of the examined Danish patients had acquired Ascaris infections from domestic pigs; ascariasis may therefore be considered a zoonotic disease in Denmark.

The prevalence and diversity of Campylobacter jejuni was investigated in pig herds on farms with and without cattle or poultry production. A bacteriological screening of pig cecal samples from 247 finisher herds was carried out at the slaughter-house. Subsequently, a follow-up study was conducted in 24 herds (either with or without prior C. jejuni isolation from pigs) in which fecal samples were collected from pigs and, if present, cattle and poultry. Samples were analyzed for presence of Campylobacter, and subsequent analysis included species identification, serotyping, and, for selected strains, pulsed-field gel electrophoresis typing. In the slaughterhouse screening, C. jejuni was isolated from pigs in 21 (8.5%) herds, but no significant difference in prevalence was found between herd types (pigs, pigs and cattle, pigs and poultry). At the slaughterhouse, C. jejuni and Campylobacter coli prevalence in pigs was 2.3 and 90.1%, respectively. In the follow-up study, herd prevalence of C. jejuni was 8.3%, whereas C. jejuni and C. coli were isolated from 0.8 and 92.0% of pigs, respectively. In mixed production herds, C. jejuni predominated in cattle (42.7%) and poultry (31.6%), whereas C. jejuni was only isolated from 1.3 to 2.5% of pigs in these herds. There were no significant differences in C. jejuni or C. coli prevalence in pigs, cattle, and poultry between herds with and without prior C. jejuni isolation at the slaughterhouse. Pulsed-field gel electrophoresis typing did not yield evidence of C. jejuni transmission between cattle or poultry and pigs in mixed production herds. In contrast, pulsed-field gel electrophoresis analysis showed indistinguishable serotypes of C. coli in pigs and cattle in two herds. Verification of C. jejuni-positive pig samples showed that individual pigs can excrete high levels of C. jejuni and that mixed infection with C. jejuni and C. coli was common in C. jejuni-positive pigs. The results of our study suggest that transmission of C. jejuni between pigs and cattle or poultry in mixed production herds occurs infrequently. Detection of indistinguishable C. coli isolates in two herds, however, might indicate the existence of low-level transmission between pigs and cattle in herds of mixed production.

This paper describes the distribution of Ascaris suum in experimentally and naturally infected pigs, and offers a comparison with A. lumbricoides infections in humans. In the first study, worms were recovered post-mortem from a group of 38 pigs that had been trickle inoculated with 10,000 infective A. suum eggs twice weekly for 12 weeks. In the second study, worms were collected from a group of 49 pigs that had been kept on a pasture contaminated with infective A. suum eggs for 10 weeks, after which they received treatment with an anthelmintic; they then were turned out on the same pasture for a second 10-week period before slaughter. The worm burdens of the naturally infected pigs were recorded both at treatment and post-mortem. Mean worm counts were similar at all occasions but the prevalence of infection was higher in the trickle infected and naturally reinfected pigs. Furthermore, the prevalence in naturally infected pigs increased significantly over the study period. Worm burden distributions in all groups were heavily overdispersed, but the distribution patterns differed significantly between groups: lower exposure (initial natural infection) gave a low prevalence and an almost uniform distribution of worm burdens among infected hosts. Continued or higher exposure (trickle and natural reinfection) resulted in increased prevalence and a reduction in the proportion of hosts with increasing worm load. A positive correlation was found between initial and reinfection worm burdens in the naturally infected pig population, suggesting that individual pigs are predisposed to a high or low intensity of infection. The prevalence and intensity as well as the distribution observed for A. suum infection in pigs were comparable to those reported for A. lumbricoides in endemic areas, and there is evidence for predisposition to A. suum in pigs, with an estimated correlation coefficient similar to that found in humans. It is concluded that A. suum infections in pigs are a suitable model to study the population dynamics of A. lumbricoides in human populations.
