Spodoptera frugiperda (J. E. Smith) (Lepidoptera: Noctuidae) is an important pest of several crops in the western hemisphere. This insect has genetically differentiated into two host-associated populations: the corn (Zea mays L.) and the rice (Oryza sativa L.) strains. The corn strain also is found in cotton (Gossypium hirsutum L.) and sorghum and the rice strain in Bermuda grass [Cynodon dactylon (L.) Pers.] and millet. In the United States and Brazil, lines from corn, rice, Bermuda grass, and millet were used to evaluate the resistance of both strains to various insecticides, and found that the corn strain is more resistant than the rice strain. However, in these studies the larvae were not genotyped. In Colombia, genotyping of fall armyworm is necessary because the rice strain also can be found in corn fields. In this work, collected larvae from corn and rice fields from Tolima (central Colombia) were genotyped and evaluated for the resistance to methomyl and lambda-cyhalothrin. We found that the rice strain does not significantly differ in resistance to methomyl compared with the corn strain but it develops tolerance more rapidly to lambda-cyhalothrin. The eggs viability of treated females also was significantly affected by methomyl on each generation. The realized heritability of resistance was higher for lambda-cyhalothrin (0.23-0.42) than for methomyl (0.04-0.14). The number of generations needed for 10-fold increase in resistance is approximately 11.5 generations for methomyl and 6.5 for lambda-cyhalothrin. Finally, the genetic basis of resistance to both insecticides involves few recessive autosomal genes. The results obtained here suggest that methomyl is a better option than lambda-cyalothin to control fall armyworm.
ABSTRAGT Spodoptera frugiperda (J.E.Smith) (Lepidoptera: Noctuidae) is a Neotropical moth that has diverged into corn. Zea mays L., and rice, Oryza sativa L., host strains because these plants are their most frequently used hosts. The corn strain also has been found in cotton, Gossypium hirsutum L., and sorghum. Sorghum bicolor (L. ) Moench, and the rice strain in small grasses and pasture grasses. Studies of the reproductive isolation between these two strains have provided ambiguous results from populations in the United States. In Golombia, we tested pre-and postzygotic isolation in these strains. Both strains showed postzygotic isolation for several life-history traits, including nutnber of egg masses, number of larvae, number of females, pupal developmental time, female and male longevity, and female and male pupal weight. We observed a reduction of the number of hybrid females and a reduction in fertility in hybrids in S. frugiperda. These results suggest tbe possibility of Haldane's lnile. Heterosis in the F^jj) and F^^) generations was observed for number of larvae and adult longevity. This line presented a high standard deviation, suggesting instability in this cross. A possible effect of the X chromosome may explain the reduction in viability and sterility in El hybrids of host strains of S. frugiperda. No temporal isolation was observed between the corn and rice strains. Differences in longevity between corn and rice strains might be another form of temporal isolation between these strains, because differences in adulthood time might reduce the encounters between them and thus hybridization.
New approaches are needed to reduce risks to the environment and natural enemies and to avoid or delay the onset of insecticide resistance. The use of insecticides based on proteinase inhibitors of hemolymph is an alternative for the control of Lepidoptera pests primarily by having low toxicity and short persistence in the environment. Thus, in this study, we describe the purification process and identification of protease inhibitors from hemolymph Anticarsia gemmatalis and their activities against trypsin enzymes. Furthermore, the three-dimensional (3D) structure of the inhibitor and binding mode to trypsin enzymes was determined, and the stability of the inhibitory activity in several pHs and temperature values was evaluated. The inhibitor was characterized as a serpin family inhibitor and named A. gemmatalis hemolymph serpin inhibitor (AHSI), with an approximate mass of 38 ± 2 kDa, highly stable to temperature and pH variations, and with inhibitory capacity on bovine trypsin and gut trypsin of A. gemmatalis demonstrated by calculated K i values and affinity energy through molecular docking, being a reversible competitive inhibitor that binds to the active site of
