Recent in vitro studies have suggested a role for sialylation in chemokine receptor binding to its ligand (Bannert, N., S. Craig, M. Farzan, D. Sogah, N.V. Santo, H. Choe, and J. Sodroski. 2001. J. Exp. Med. 194:1661–1673). This prompted us to investigate chemokine-induced leukocyte adhesion in inflamed cremaster muscle venules of α2,3 sialyltransferase (ST3Gal-IV)-deficient mice. We found a marked reduction in leukocyte adhesion to inflamed microvessels upon injection of the CXCR2 ligands CXCL1 (keratinocyte-derived chemokine) or CXCL8 (interleukin 8). In addition, extravasation of ST3Gal-IV−/− neutrophils into thioglycollate-pretreated peritoneal cavities was significantly decreased. In vitro assays revealed that CXCL8 binding to isolated ST3Gal-IV−/− neutrophils was markedly impaired. Furthermore, CXCL1-mediated adhesion of ST3Gal-IV−/− leukocytes at physiological flow conditions, as well as transendothelial migration of ST3Gal-IV−/− leukocytes in response to CXCL1, was significantly reduced. In human neutrophils, enzymatic desialylation decreased binding of CXCR2 ligands to the neutrophil surface and diminished neutrophil degranulation in response to these chemokines. In addition, binding of α2,3-linked sialic acid–specific Maackia amurensis lectin II to purified CXCR2 from neuraminidase-treated CXCR2-transfected HEK293 cells was markedly impaired. Collectively, we provide substantial evidence that sialylation by ST3Gal-IV significantly contributes to CXCR2-mediated leukocyte adhesion during inflammation in vivo.
L‐selectin belongs to the C‐type lectin family of glycoproteins and is constitutively expressed on most leukocytes. L‐selectin mediates leukocyte rolling in inflamed microvessels and high endothelial venules (HEV) via binding to specific carbohydrate structures on selectin ligands. Previous studies using sialidase treatment suggested a role of sialic acid residues in L‐selectin‐dependent rolling. To investigate the role of the α2,3‐sialyltransferase (ST3Gal)‐IV on L‐selectin ligand activity in vivo, we studied leukocyte rolling in inflamed venules of the cremaster muscle and in Peyer's patch HEV of ST3Gal‐IV‐deficient mice and littermate control mice. In cremaster muscle venules with or without TNF‐α treatment, L‐selectin‐dependent rolling was almost completely abolished in ST3Gal‐IV–/– mice. In both models, L‐selectin interacts with P‐selectin glycoprotein ligand‐1 (PSGL‐1) presented by adherent leukocytes and leukocyte fragments, but not with endothelial L‐selectin ligands. In contrast, L‐selectin‐dependent rolling in Peyer's patch HEV, which is mediated by unknown endothelial L‐selectin ligands, was not impaired in the absence of ST3Gal‐IV. Our in vivo data show that PSGL‐1, the molecule responsible for L‐selectin‐mediated leukocyte interactions in inflammation, is dependent on ST3Gal‐IV, while α2,3‐sialylation by ST3Gal‐IV is not necessary for L‐selectin ligand activity on high endothelial cells of Peyer's patch HEV.
52Известно, что при стойкой артериальной гипер-тонии (АГ) одной из ведущих компенсаторных реак-ций, обусловленных системной гипоксией, является усиление эритропоэза. В этой ситуации на фоне повышенного периферического сосудистого сопро-тивления происходит ухудшение реологических свойств крови, связанное с повышением гемато-крита и вязкости крови. С другой стороны, течение гипоксического синдрома может осложняться в связи с несостоятельностью определённого пула эритроцитов проникать в сосуды микроциркулятор-ного русла для осуществления эффективного газо-обмена. Такое явление, несомненно, связано с пато-логическими изменениями в самой мембране этих клеток. Изменения цитоскелета эритроцитов у боль-ных АГ может проявляться в различных формах отклонения от нормальных клеток, в том числе и в виде сфероцитоза [1] Цель. Исследовать связь между характером изменения показателя сферич-ности эритроцитов и уровнем структурно-функциональных белков мембраны эритроцитов у больных артериальной гипертонией (АГ). Материал и методы. В исследовании участвовали больные АГ I и II степени (всего 51, средний возраст -42±1,5 лет), у которых определяли показатель сферичности эритроцитов (ПСЭ) и проводили количественную оценку 10-ти мембранных белков эритроцитов. Анализ полученных данных проводили методами непараметрической и параметрической статистики. Результаты. Все больные АГ были разделены на группу с "нормоцитозом" (ПСЭ ≥3,4 усл. ед.) и группу со "сфероцитозом" (ПСЭ <3,4 усл. ед.), которые достоверно отличались по содержанию α-и β-спектринов. Дискриминантный анализ выявил, что наибольший вклад в разделение этих 2-х групп внесли следующие белки: α-спектрин, АТБ, тропомиозин и Г-3-ФДГ. Те же самые белки были выявлены как предикторы отклонения ПСЭ в модельном плане при регрессионном анализе. Заключение. Было показано, что нарушение количества и соотношения раз-личных как цитоскелетных, так и функциональных белков в мембране эритро-цитов при АГ может обусловливать изменение стабильности и деформабель-ности самой мембраны, что, в свою очередь, будет содействовать развитию существенной дисфункции эритроцитов в целом и осложнять течение систем-ной гипоксии у данной категории больных. Aim. To study the relation of the pattern of erythrocyte sphericity value changes and the levels of structural and functional proteins of erythrocyte membrane in patients with arterial hypertension (AH). Material and methods. Totally, 51 patient participated with AH of 1 and 2 grade (mean age 42±1,5 y.), in whom we assessed sphericity of erythrocytes (SE) and performed quantitative evaluation of 10 membrane proteins of erythrocytes. Analysis of the acquired data was done via non-and parametrical statistics methods. Results. All AH patients were selected to a group with "normocytosis" (SE ≥3,4 U) and "spherocytosis" (SE <3,4 U), that significantly differed by contents of α-and β-spectrines.
