Fibrosis is a pathological feature observed in patients with Duchenne muscular dystrophy (DMD) and in mdx mice, the experimental model of DMD. We evaluated the effect of suramin, a transforming growth factor-beta 1 (TGF-β1) blocker, on fibrosis in mdx mice. mdx mice (6 months old) received suramin for 7 weeks. Suramin- and saline-treated (control) mdx mice performed exercise on a treadmill to worsen disease progression. Immunoblotting showed an increase of TGF-β1 in mdx diaphragm, limb, and cardiac muscles. Suramin decreased creatine kinase in mdx mice and attenuated fibrosis in all muscles studied, except for cardiac muscle. Suramin protected limb muscles against damage and reduced the exercise-induced loss of strength over time. These findings support a role for TGF-β1 in fibrinogenesis and myonecrosis during the later stages of disease in mdx mice. Suramin might be a useful therapeutic alternative for the treatment of dystrophinopathies.

One dose of LAIV provided clinically significant protection against influenza in young children previously unvaccinated against influenza; 2 doses provided additional protection. Protection after 2 doses in year 1 persisted through a second season without revaccination. LAIV excipients were not a major contributor to reactogenicity. These benefits provide support for increased use of LAIV in children > or =2 years of age.

Intrinsic laryngeal muscles share many anatomical and physiological properties with extraocular muscles, which are unaffected in both Duchenne muscular dystrophy and mdx mice. We hypothesized that intrinsic laryngeal muscles are spared from myonecrosis in mdx mice and may serve as an additional tool to understand the mechanisms of muscle sparing in dystrophinopathy. Intrinsic laryngeal muscles and tibialis anterior (TA) muscle of adult and aged mdx and control C57Bl/10 mice were investigated. The percentage of central nucleated fibers, as a sign of muscle fibers that had undergone injury and regeneration, and myofiber labeling with Evans blue dye, as a marker of myofiber damage, were studied. Except for the cricothyroid muscle, none of the intrinsic laryngeal muscles from adult and old mdx mice showed signs of myofiber damage or Evans blue dye labeling, and all appeared to be normal. Central nucleation was readily visible in the TA of the same mdx mice. A significant increase in the percentage of central nucleated fibers was observed in adult cricothyroid muscle compared to the other intrinsic laryngeal muscles, which worsened with age. Thus, we have shown that the intrinsic laryngeal muscles are spared from the lack of dystrophin and may serve as a useful model to study the mechanisms of muscle sparing in dystrophinopathy.

This study reports an investigation of the pharmacological activity, cytotoxicity and local effects of a liposomal formulation of the novel local anaesthetic ropivacaine (RVC) compared with its plain solution. RVC was encapsulated into large unilamellar vesicles (LUVs) composed of egg phosphatidylcholine, cholesterol and alpha-tocopherol (4:3:0.07, mole %). Particle size, partition coefficient determination and in-vitro release studies were used to characterize the encapsulation process. Cytotoxicity was evaluated by the tetrazolium reduction test using sciatic nerve Schwann cells in culture. Local anaesthetic activity was assessed by mouse sciatic and rat infraorbital nerve blockades. Histological analysis was performed to verify the myotoxic effects evoked by RVC formulations. Plain (RVC(PLAIN)) and liposomal RVC (RVC(LUV)) samples were tested at 0.125%, 0.25% and 0.5% concentrations. Vesicle size distribution showed liposomal populations of 370 and 130 nm (85 and 15%, respectively), without changes after RVC encapsulation. The partition coefficient value was 132 +/- 26 and in-vitro release assays revealed a decrease in RVC release rate (1.5 fold, P < 0.001) from liposomes. RVC(LUV) presented reduced cytotoxicity (P < 0.001) when compared with RVC(PLAIN). Treatment with RVC(LUV) increased the duration (P < 0.001) and intensity of the analgesic effects either on sciatic nerve blockade (1.4-1.6 fold) and infraorbital nerve blockade tests (1.5 fold), in relation to RVC(PLAIN). Regarding histological analysis, no morphological tissue changes were detected in the area of injection and sparse inflammatory cells were observed in only one of the animals treated with RVC(PLAIN) or RVC(luv) at 0.5%. Despite the differences between these preclinical studies and clinical conditions, we suggest RVC(LUV) as a potential new formulation, since RVC is a new and safe local anaesthetic agent.

Stretch-activated channels (SACs) might be directly involved in the pathology of DMD, and transient receptor potential cation channels have been proposed as likely candidates of SACs. We investigated the levels of transient receptor potential canonical channel 1 (TRPC1) and the effects of streptomycin, a SAC blocker, in muscles showing different degrees of the dystrophic phenotype. Mdx mice (18 days old, n ϭ 16) received daily intraperitoneal injections of streptomycin (182 mg/kg body wt) for 18 days, followed by removal of the diaphragm, sternomastoid (STN), biceps brachii, and tibialis anterior muscles. Control mdx mice (n ϭ 37) were injected with saline. Western blot analysis showed higher levels of TRPC1 in diaphragm muscle compared with STN and limb muscles. Streptomycin reduced creatine kinase and prevented exercise-induced increases of total calcium and Evans blue dye uptake in diaphragm and in STN muscles. It is suggested that different levels of the stretch-activated calcium channel protein TRPC1 may contribute to the different degrees of the dystrophic phenotype seen in mdx mice. Resumo -Setenta e cinco amostras de madeiras de clones de Eucalyptus, obtidas de diferentes regiões geográficas do Brasil, foram utilizadas para este estudo. Análises estatísticas demonstraram que a carga de álcali ativo para polpação, o teor de lignina e o teor de extrativos apresentaram efeitos significativos no rendimento da polpação kraft. A densidade básica da madeira não afetou significativamente o rendimento da polpação, mas sim o consumo de madeira para produção de celulose (m 3 madeira/t celulose). Densidade básica, teor de lignina e teor de extrativos foram recomendados como critérios de qualidade da madeira a serem determinados para seleção de clones antes da determinação das características de polpação.Palavras-chave: Eucalyptus, Qualidade da madeira e Rendimento kraft. ANALYSIS OF WOOD QUALITY CRITERIA OF EUCALYPTUS WOOD FOR KRAFT PULP PRODUCTION INTRODUÇÃOO Brasil ocupa, atualmente, a quarta posição entre os maiores produtores mundiais de celulose. Segundo dados da Bracelpa, referentes a 2008, a produção nacional de celulose, incluindo as pastas de alto rendimento, atingiu nesse ano a cifra de 12,7 milhões de toneladas. Dessa produção, mais de 96% foi de polpa celulósica química. A produção nacional de polpa química foi alcançada predominantemente com madeiras de eucalipto que representaram mais de 86% da produção total, tendo as madeiras de Pinus sido responsáveis pelos restantes 14%. O formidável crescimento da indústria nacional de celulose deverá continuar nos próximos anos e existem previsões de que em 2012 a produção nacional de celulose deverá ultrapassar 14 milhões de toneladas, posicionando o Brasil como o quarto maior produtor mundial de celulose.O Brasil apresentou, nas últimas décadas, uma invejável evolução na produtividade das florestas de Eucalyptus, passando de 15 m 3 /ha/ano na década de 1970 para, atualmente, uma média nacional de c e r c a d e 4 5 m 3 / h a / a n o . E s t u d o r e c e n t e d e caracterização dos melhores clones de Eucalyptus cultivados no Brasil (GOMIDE et al., 2005), envolvendo as principais indústrias nacionais de celulose, demonstrou que esses plantios florestais no Brasil apresentam os mais elevados níveis mundiais de incremento médio anual (IMA). Os clones de Eucalyptus analisados neste estudo demonstram elevada produtividade, tendo 20% dos clones atingido IMA igual ou superior a 50 m 3 /ha/ano, e 70% dos clones apresentaram incrementos acima de 40 m 3 /ha/ano.
