When activity of peroxidase in adult Paragonimus westermani was monitored using o-dianisidine and H2O2 as substrates, its specific activity was 1.5 times higher in excretory-secretory product (ESP) than in crude extract. The enzyme was purified by two purification steps of Sephacryl S-300 Superfine gel permeation and DEAE-Trisacryl M anion exchange chromatographies. Its activity increased 16.9 fold with 32.3% recovery. The enzyme was inhibited totally by 1 millimoles of dithiothreitol (DTT), 2-mercaptoethanol and azide. Molecular mass was 16 kDa in reducing SDS-polyacrylamide gel electrophoresis (PAGE) or 19 kDa in TSK-Blue gel filtration high performance liquid chromatography (HPLC), respectively. Special staining for peroxidase by diaminobenzidine on SDS-PAGE confirmed the activity. The peroxidase was less reactive to a paragonimiasis serum when observed by SDS-PAGE/immunoblot. In addition, specific activities of superoxide dismutase (SOD) and catalase were also identified in the ESP. High activities of these antioxidant enzymes in ESP indicate that they are parts of defense mechanisms against reactive oxygen intermediates from host.
− Metallic sintered brake pads are often applied to mid/high speed train due to their high strength and thermal characteristics. Imbalance contact between discs and pads can greatly influence the life span, one sided wear, discs attack/crack and threat the safety of the train during operation. In this research, we analyzed pressure/ temperature distribution between brake pads and disks. Analyzed data had been verified and modified to conduct further tests of flexible brake pads with small/full-scale dynamo test. Flexible brake pads were installed to high speed train to conduct further tests to identify the differences between rigid brake pads and flexible brake pads. In result, Flexible brake pads showed outstanding disk thermal stability, one sided wear, noise and wear rate than rigid brake pad.
An important problem in reliability analysis for repairable systems is to model the maintenance effect. The most of researches have assumed two extreme cases; one is perfect maintenance and the other is minimal maintenance. However, many of maintenances performed by domestic subway operators are imperfect maintenances which have the effect between both of two extreme cases. This article deals with the problem determining the imperfect preventive maintenance intervals based on failure data in units of the PDS(passenger door system) in Metro EMU. This paper deals with a case study on determining imperfect maintenance interval by using the level of maintenance effect through reliability analysis of PDS.
The LRT(light rail transit) system, which has medium transport capacity between subway and bus is an advanced transportation system. It has many benefits like chap construction and operational cost down through driverless and flexible route planning. The rubber tired AGT(K-AGT) is a kind of LRT, which has rubber wheels and side guides. The side guides has many advantages, but brings vibration and noise problems from the friction between the guide rail and the wheels. This is the major source for the of passengers' discomfort. The purpose of this paper is to analyze dynamic characteries and running stability of the rubber tired AGT localization bogie if the AGT's speed is increased from 70 km/h to 80 km/h. The current design parameters of bogie suspension, as it is designed, was examined to satisfy the comfort index of the railway vehicle in performance test.
