Populations of large and small milk fat globules were isolated and analyzed to determine differences in fatty acid composition. Globule samples were obtained by centrifugation from milks of a herd and of individual animals produced under both pasture and barn feeding. Triacylglycerols of total globule lipids were prepared by thin layer chromatography and analyzed for fatty acid composition by gas chromatography. Using content of the acids in large globules as 100%, small globules contained fewer short-chain acids, -5.9%, less stearic acid, -22.7%, and more oleic acids, +4.6%, mean values for five trials. These differences are consistent with alternative use of short-chain acids or oleic acid converted from stearic acid to maintain liquidity at body temperature of milk fat globules and their precursors, intracellular lipid droplets. Stearyl-CoA desaturase (EC 1.14.99.5), which maintains fluidity of cellular endoplasmic reticulum membrane, is suggested to play a key role in regulating globule fat liquidity. Possible origins of differences between individual globules in fatty acid composition of their triacylglycerols are discussed.
Die Milchfettzusammensetzung wird in starkem Maße von den Fütterungsbedingungen beeinflußt. Dies kann beträchtliche Auswirkungen auf die Qualität vieler Milchprodukte haben. Um die möglichen Variationen der physikalischen und chemischen Eigenschaften des Milchfettes als Folge des ernährungsphysiologischen Zustandes der Kuh zu erfassen, wurde eine kleine Kuhherde nacheinander definierten Fütterungsbedingungen unterworfen (Stall‐, Weide‐ und energetische Unterfütterung). Die Untersuchungen zeigten, daß von der Stall‐ über die Weidefütterung bis zum Energiedefizit der Anteil an Ölsäure sowie langkettigen Triglyceriden ansteigt, während der Anteil kurz‐ bis mittelkettiger Fettsäuren und Triglyceride sich verringert. Trotz unterschiedlichster Fettzusammensetzungen ergeben sich relativ konstante Werte für die Viskosität und das mittlere Triglyceridmolekulargewicht; darüber hinaus stimmen verschiedene Charakteristika der Schmelz‐ und Erstarrungskurven nahezu überein. Es erfolgt eine zunehmende Angleichung der Fest‐/Flüssig‐Verhältnisse bei höheren Temperaturen. Diese Ergebnisse weisen auf Regulationsmechanismen hin, die dadurch gekennzeichnet sind, daß durch die Gegenläufigkeit der Anteile kurzkettiger Fettsäuren und der Ölsäure die Fließ‐ und Schmelzeigenschaften in physiologisch erforderlichen engen Bereichen gehalten werden. Bei Weide‐ gegen‐über Stallfütterung nehmen alle Isomeren der Ölsäure sowie die Linolsäure zu, beim Energiedefizit liegen trotz hohen Ölsäuregehaltes die entsprechenden Anteile auf dem niedrigen Niveau der Stallfütterung. Diese Ergebnisse sowie die kontinuierliche Abnahme des Cholesteringehaltes von Stall‐ über Weide‐ bis hin zur Unterfütterung werden an Hand der unterschiedlichen Fettstoffwechselwege erklärt; des weiteren werden die trans‐C18: 1‐Anteile im Zusammenhang mit der Höhe des Milchfettgehaltes diskutiert. Die Fettkügelchen sind während der Unterfütterungsperiode besonders groß und instabil. Als Folge sind Nachteile für Schlagrahm (Aufrahmung, Pfropfenbildung) und andere Milchprodukte (Lipolyse) zu erwarten.
