A major antibody combining site on foot and mouth disease virus (FMDV) serotype O1K has been identified in a predicted surface helix of viral protein 1 (VP1) between amino acid residues 144 and 159. A hexadecapeptide covering this sequence elicits high titers of antibodies that specifically recognize and neutralize FMDV. The high quality of the immune response is attributed to a particularly stable conformation of the antigenic amino acid sequence, which is most likely an alpha‐helix.
SUMMARYThe labelled RNAs of the three European types of foot-and-mouth disease virus (A2-sVAIN ; C-OBERBAYERN; O ~-LOMBARDY) have been cross-hybridized with a large excess of denatured unlabelled foot-and-mouth disease virus-specific doublestranded RNA of the respective virus types. The degree of double-strand reformation in the heterologous reactions was taken as a measure of the extent to which polynucleotide sequences were shared by the virus RNAs. Different degrees of hybridization were obtained across different pairs of virus types: A2-SPAIN with O I-LOMBARDY = 65 %, A2-SPAIN with C-OBERBAYERN = 44 Yo, O I-LOMBARDY with C-OBERBAYERN = 45 %.
ZusammenfassungPferdearteriitisvirus (EAV) Stature Bucyrus wurde in BHK 21-Zellkulturen gezfichtet und die extra-und intrazellul~re Virusvermehrung ermittelt.Die l~lotationsdichte des Virus betr~gt 1,20 g/cm a. Nach Konzentrierung durch Ultrazentrifugierung und partieller Reinigung durch Dichte-und Zonenzentrifugierung im Saccharosegradienten konnten elektronenoptisch Viruspartikel eines Durchmessers yon 60 • 13 m~ nachgewiesen werden. Die Viria bestehen aus einer Hiille ohne Projektionen und einem Innen-kSrper yon 35 • 9 m~.Die taxonomische Klassifikation des Virus wird diskutiert. Summary Equine Arteritis Virus: Multiplication in BHK 21-Cells, Buoyant Density and Electron Microseopical Demonstration Equine Arteritis Virus (EAV) strain Bueyrus was multiplied in BHK21-cells; a growth curve considering extra-and intraeellular virus was established.The buoyant density of EAV in sucrose gradients was 1.20 g/ml. After concentration of virus material by ultracentrifugation and partial purification by isopycnic and rate zonal centrifugations in sucrose gradients virus particles of 60=[= 13mfz, Size were demonstrated electron microscopically. The viria consist of an envelope without projections and an internal core of 35 -4-9 m~z in diameter.The results are discussed with respect to virus classification.
Zusammenfassung Bei der Untersuchung natürlich erkrankter und experimentell infizierter (immuner) Rinder wurde MKS‐Dauerausscheidervirus, Typ O, im Ösophageal‐Pharyngealgeschabsel über einen Zeitraum von 9 Monaten nachgewiesen. Die erhobenen Befunde sprechen für ein intrazelluläres Persistieren des Virus in bestimmten Schleimhautbezirken des Ösophagus und des Retropharyngealraumes. Die isolierten Dauerausscheidervirusstämme hatten eine erhöhte Interferonproduktion und ‐empfindlichkeit, eine verminderte zytopathogene Wirkung für Gewebekulturen, eine verlängerte Inkubationszeit für Säuglingsmäuse und sie bildeten kleine Plaquevarianten, so daß zu vermuten ist, daß es sich bei den aus Dauerausscheiderrindern isolierten Stämmen um modifiziertes MKS‐Virus handelt, das zusätzlich durch Speichelantikörper in seiner Pathogenität gehemmt wird. Anhand von Ergebnissen der Pathogenitäts‐ und Kontaktübertragungsversuche bei Rindern und Schweinen wird die epizootiologische Bedeutung des MKS‐Dauerausscheidervirus diskutiert. Summary Studies on permanent virus excretors in cattle vaccinated and experimentally infected with foot‐and‐mouth (FM) disease Foot‐and‐mouth disease virus type O was recovered from oesophageal/pharyngeal scrapings of cattle up to nine months after natural and experimental infection with the virus. This suggests intracellular persistence of the virus in certain areas of the mucosa of the oesophagus and retro‐pharyngeal area. The strains of persistent virus which were isolated showed an increased production of and susceptibility to interferon, a prolonged incubation period in suckling mice and produced small plaque variants, all of which findings suggest that the strains of virus isolated from cattle that have become permanent excretors represent a modified FM virus which has been inhibited in its pathogenicity by salivary antibodies. The epidemiological significance of permanent excretor virus is discussed on the basis of results from pathogenicity and contact transfer experiments in cattle and pigs. Résumé Étude des porteurs du virus aphteux persistant sur des bovins vaccinés et expérimentalement infectés Virus aphteux, typ O, fut isolé pendant 9 mois dans le liquide oesophageal et retropharyngeal des bovins après l'infection naturelle et expérimentale. Ces résultats prouvent que ce virus persiste intracellulaire dans certains districts de la muqueuse oesophageal et retropharyngeal. Les souches isolées du virus aphteux persistant montraient une production et une sensibilité élevée à l'interféron, une action cytopathogène affaiblie pour cellules tissulaires, une période d'incubation prolongée pour souriceaux nouveau‐nés et elles formaient petites plaques. De ce fait on peut présumer qu'il s'agit d'un virus aphteux modifié qui est en outre bloqué par anticorps antiaphteux dans la salive. La conséquence epizootiologique de ce virus est discutée en vue des résultats experimentales obtenus après contact avec bovins et porcins réceptives.
