The venom of amphibians is a fascinating source of active substances. In view of their medical importance and aiming to explore the amazing Brazilian biodiversity, we conducted bioprospecting of antiproliferative activity in extracts of Rhinella marina and Rhaebo guttatus toads occurring in the Southern Amazon of Mato Grosso, Brazil. LC-MS and HPLC analysis of the venom extracts of R. marina revealed four bufadienolides (telocinobufagin, marinobufagin, bufalin and resibufogenin. R. guttatus venom extracts contained only marinobufagin. First, R. marina and R. guttatus venom extracts were evaluated for cytotoxicity against tumor cell lines by the MTT assay. All extracts revealed cytotoxicity, where R. marina extracts were comparable to doxorubicin (IC₅₀ values ranging from 0.01 to 0.23 μg/mL). Only extracts of R. guttatus toad venom caused membrane disruption of human erythrocytes. The extracts were investigated for selective activity by determining their effect on stimulated human peripheral blood mononuclear cells (PBMC) with the Alamar Blue™ assay. The extracts were up to 80-fold more selective against leukemia cells when compared to dividing leukocytes. Aiming to confirm these antiproliferative effects, BrdU incorporation into DNA was measured in HL-60 treated cells with R. marina venom extracts. These extracts decreased BrdU incorporation at both concentrations tested. In summary, nine extracts of R. marina and R. guttatus venom showed pronounced lethal and discriminating effects on tumor lines, especially those from R. marina, highlighting toad parotoid gland secretions as a promising source for novel lead anticancer chemicals.

Phytochemical investigation of ethanolic leaves extracts of T. fagifolia led to the isolation of (+)-catechin, sitosterol-3-O-β-D-glucopyranoside, α-and β-tocopherol, a mixture of lupeol, α-and β-amyrin, sitosterol and a mixture of glicosid flavonoids (CP-13). The structures of these compounds were identified by 1 H and 13 C NMR spectral analysis and comparison with literature data. Absolute configuration of the catechin was determinate by circular dichroism. Antioxidant activity (EC 50 ), evaluated by 2,2-diphenyl-1-picrylhidrazyl (DPPH) assay system, decreased in the order: (+)-catechin > hydroalcoholic fraction > CP-13 > aqueous fraction > EtOH extract.Keywords: Terminalia fagifolia; DPPH, antioxidant activity. INTRODUÇÃOA família Combretaceae compreende 18 gêneros, sendo Combretum e Terminalia os mais abundantes, com cerca de 370 e 200 espé-cies, respectivamente, as quais são largamente distribuídas no Oeste e Sul da África e amplamente utilizadas na medicina popular. 1As plantas do gênero Terminalia são ricas em triterpenos pentací-clicos e seus derivados glicosilados, flavonoides, taninos e outros compostos aromáticos.2 Apresentam diversas atividades farmacológicas, tais como antifúngica, anti-helmíntica, antimalárica, anticancerígena, hipoglicêmica, anti-inflamatória, antibacteriana, antioxidante, antiulcerogênica, antiviral, antidepressora, tripanocida, moluscicida, imunomodulatória e efeitos hepatoprotetor, cardioprotetor, dentre outros. Terminalia fagifolia Mart. et Zucc é uma planta encontrada no cerrado, distribuída pela Bahia, Ceará, Goiás, Distrito Federal, Piauí, São Paulo, Maranhão, Mato Grosso, Mato Grosso do Sul e Minas Gerais. É conhecida popularmente como capitão, capitão-do-mato, capitão-docerrado, capitão-do-campo, mirindiba e pau-de-bicho. A árvore é melífera e ornamental, sendo seus frutos utilizados no artesanato e a madeira pode ser empregada na marcenaria e construção civil. Na medicina popular a casca do caule é usada no combate a aftas e tumores. 4Estudos realizados com T. fagifolia demonstraram que o extrato etanólico das cascas apresenta atividades citotóxica e antioxidante, bem como a ocorrência de flavonoides (flavanonas, chalconas e flavanas), 1,3-diarilpropanos, triterpenos pentacíclicos glicosilados e não-glicosilados e um esteroide, o sitosterol.5 O extrato etanólico das folhas apresenta forte potencial antioxidante no ensaio do DPPH. 6 Nos últimos anos, um grande interesse no estudo de antioxidantes tem ocorrido devido, principalmente, às descobertas sobre o efeito dos radicais livres no organismo. The drug-resistance of malaria parasites is the main problem in the disease control. The huge Brazilian biodiversity promotes the search for new compounds, where the animal kingdom is proving to be a promising source of bioactive compounds. The main objective of this study was to evaluate the antiplasmodial and cytotoxic activity of the compounds obtained from the toad venoms of Brazilian Amazon. Toad venoms were collected from the secretion of Rhinella marina and Rhaebo guttatus in Mato Grosso State, Brazil. The powder was extracted at room temperature, yielding 2 extracts (RG and RM) and a substance (‘1’) identified as a bufadienolide, named telocinobufagin. Growth inhibition, intraerythrocytic development, and parasite morphology were evaluated in culture by microscopic observations of Giemsa-stained thin blood films. Cytotoxicity was determined against HepG2 and BGM cells by MTT and neutral red assays. The 2 extracts and the pure substance (‘1’) tested were active against chloroquine-resistant Plasmodium falciparum strain, demonstrating lower IC50 values. In cytotoxic tests, the 2 extracts and substance ‘1’ showed pronounced lethal effects on chloroquine-resistant P. faciparum strain and low cytotoxic effect, highlighting toad parotoid gland secretions as a promising source of novel lead antiplasmodial compounds.

THE Protium heptaphyllum RESIN: ISOLATION, STRUCTURAL CHARACTERIZATION AND EVALUATION OF THERMAL PROPERTIES. Three mixtures of triterpenes (maniladiol and breine; α and β-amyrin; lupenone, α and β-amyrinone) were isolated from Protium heptaphyllum March resin. The structural identification was based on NMR and mass spectrometry data. Lupenone, and α and β-amyrinone were not reported before as constituents of this resin. The resin was submitted to methylation and acetylation reactions. The pure and derivatized resins and the mixtures (maniladiol and breine; α and β-amyrin) were analyzed by TG and DSC. The TG curves revealed that the derivatization decreases the thermal stability of the resin. The DSC curves showed peaks that can be assigned to evaporation and phase transitions processes.Keywords: Burseraceae; triterpenes; thermal analysis. INTRODUÇÃOO estudo de produtos naturais têm avançado bastante nos últi-mos tempos em função do desenvolvimento de novas técnicas espectroscópicas e cromatográficas. O isolamento dessas substân-cias naturais fornece matéria-prima importante para o preparo de diversos produtos industrializados, o que requer o conhecimento de suas propriedades físicas e químicas. As propriedades térmicas das substâncias, tais como as transições de fase e processos de degradação térmica, têm um papel decisivo na qualidade do produto final, podendo ser convenientemente estudadas por termogravimetria (TG) e calorimetria exploratória diferencial (DSC) 1 . A espécie Protium heptaphyllum March (Burseraceae), conhecida popularmente como almécega, é encontrada na região Amazônica, no Piauí, na Bahia, em Minas Gerais e Goiás, e em países como Suriname, Colômbia, Venezuela e Paraguai 2 . Esta espécie exsuda uma resina oleosa e amorfa, cujas aplicações gerais vão desde a fabricação de vernizes e tintas, na calafetagem de embarcações, em cosméticos e em repelentes de insetos. Apresenta algumas indicações terapêuticas, como cicatrizante e expectorante e as ações antiulcerogênica e antiinflamatória já comprovadas 3,4 . A resina da almécega é constituída por substâncias de natureza terpênica, sendo o óleo essencial rico em monoterpenóides e fenilpropanóides 4,5 . Entre os constituintes fixos, a literatura registra a presença de um monoterpeno trioxigenado e quatro misturas biná-rias de triterpenóides 3,6 . No presente trabalho, foram isoladas três misturas, totalizando sete triterpenos pentacíclicos (Figura 1), da resina de almécega, identificados como α-amirina (1a), β-amirina (2a), breína (3a), maniladiol (4a), α-amirinona (5a), β-amirinona (6a) e lupenona (1b). Reações de acetilação e metilação foram efetuadas com a resina, devido à presença de grupamentos alcóolicos e ácidos em seus constituintes. Curvas termogravimétricas (TG) e de calorimetria exploratória diferencial (DSC) foram obtidas para a resina natural, derivatizada, e para os principais constituintes, com o objetivo de avaliar suas propriedades térmicas e contribuir para a busca de possíveis aplicações tecnológicas deste material.
