The Methods based on genomic and proteomic approaches are described as effective tools for the identification of microorganisms. The development of methodologies capable of differentiating, interspecifically, pathogenic, wild and genetically modified Escherichia coli strains is desirable for the fields of healthcare and Biotechnology. The purpose of this study was to verify the viability of ERIC-PCR and MALDI TOF methods in differentiating lineages of Escherichia strains. For this purpose, laboratory Escherichia coli ATCC8739, Escherichia coli W3110, Escherichia coli BL21DE3+, Escherichia coli JM109, Escherichia coli MC 1061 and Escherichia coli DH5α were subjected to ERIC-PCR and MALDI TOF mass spectrometry analyzes. Genomic (ERIC-PCR) and proteomic (MALDI-TOF) methods were able to discriminate between different lineages of Escherichia coli strains including lineages of Escherichia coli K-12. However, the MALDI TOF proteomic approach revealed being able to differentiate interspecifically lineages of Escherichia coli strains. The determination of the most frequent masses found in the studied Escherichia coli strains in addition to future experiments of peptide sequencing profile and SDS-PAGE can be used as a guideline for validating a method for proteomic identification of these strains.
Inovações em metodologias para identificação de microrganismos genômicas e proteômicas associadas a tecnologias digitais estão em alinhamento com a visão de indústria 4.0 e estão em ascensão. O objetivo desse trabalho foi o desenvolvimento de um protótipo de um aplicativo (App) para a identificação de fungos filamentosos e leveduras ao nível de espécie. A construção do protótipo foi realizada de modo a apresentar uma aplicação web com interface responsiva. O App foi desenvolvido em processo cloud computing com modelo em cascata. Como parte dos requisitos do App foi construído um banco de dados Cloud Firestore com processamento de imagens através de uma biblioteca skImage. Para tal, foram selecionados géis de agarose com perfis de restrição de fungos filamentosos e leveduras previamente identificados ao nível de espécie por metodologias genômicas (PCR/RFLP) e proteômica (espectrometria de massa). O App identificado como iGENE foi capaz de realizar o reconhecimento de perfis de restrição de géis de agarose, comparando-o aos fungos filamentosos e leveduras cadastrados em sua biblioteca. O resultado ao nível de espécie foi possível para perfis com similaridade superior a 90%. Embora as imagens analisadas tenham apresentado esse perfil, o App foi construído de modo a considerar também identificações ao nível de gênero para similaridades entre 89 e 70%, bem como “microrganismo não identificado” abaixo desse escore. A inclusão de novas espécies de fungos filamentosos e leveduras na biblioteca do App permitirá uma maior robustez na geração do resultado da identificação ao nível de espécie.
Introdução: O combate à pandemia da COVID-19 decretada pela Organização Mundial da Saúde (OMS) tem impulsionado pesquisas científicas em todo o mundo, como ou entre eles, a caracterização de aspectos da genômica e proteômica relacionados ao SARS-CoV-2. O SARS-CoV-2 é responsável por ocasionar doenças respiratórias e entéricas, sendo associado às infecções agudas e graves do trato respiratório e tem mostrado um impacto significativo na saúde humana global. A análise das ômicas fornece informações importantes da evolução do vírus, um dado importante para o desenvolvimento de vacinas eficazes. Métodos: Este artigo foi desenvolvido utilizando-se o método de revisão integrativa a partir de buscas na literatura nos seguintes bancos de dados: PubMed, site de buscas mantido pela Biblioteca Nacional de Medicina (NLM ®) dos Estados Unidos; na PeerJ, revista científica de acesso aberto com revisão por pares e na Science Direct, plataforma para acesso a revistas científicas. Resultados: Os resultados foram compostos por onze artigos científicos, selecionados pelos critérios de inclusão previamente estabelecidos. Destes, oito foram encontrados na base de dados PubMed, um na PeerJ e dois na Science Direct. Conclusões: As variações relatadas do vírus, indicam o grande potencial de ocorrência de mutação através de exclusões e recombinações genômicas, as quais são fatores relevantes e que podem gerar desafios para o desenvolvimento de vacina.
