Paclobutrazol PBZ , [ RS, RS]--[ -chlorophenyl]-, -dimethyl--H-, , -triazol--yl pentan--ol, consists of a triazole ring and a benzene ring-chloro linked to a carbon chain open. It is a plant growth regulator widely used in many crops in order to produce fruit throughout the year by inhibiting gibberellin synthesis, a hormone responsible for the vegetative plant growth. Actually, studies are showing that paclobutrazol remains active in the soil for a long time, affecting the growth and development of subsequent crops by reducing plant vigor. Biodegradation is an effective and cheap process that can to degrade or transform contaminants to less toxic or nontoxic. In this work, the biodegradation of paclobutrazol was studied using in submersed culture and saturated and unsaturated soils. In these conditions, experiments with biostimulation and bioaugmentation were performed. In the experiments carried out in submersed culture, with biostimulation by addition of glycerol, the PBZ biodegradation was higher than that with PBZ as sole carbon source. The biodegradation of PBZ in unsaturated soils was more efficient when soil samples with a history of application of PBZ were used. The highest number of applications of PBZ favored biodegradation. The objective of this study was to investigate the paclobutrazol biodegradation in two soils from the São Francisco River Valley. The biodegradation experiments were conducted in batch using paclobutrazol and paclobutrazol added glycerol. The experiments were performed in sterile and nonsterile conditions using a mixed culture of Pseudomonas. The concentration of paclobutrazol was determined by high performance liquid chromatography. The biodegradation reached 43% in 14 days of experiments with only paclobutrazol and 70% in 28 days of experiments that contained glycerol and paclobutrazol.Keywords: biodegradation; paclobutrazol; Pseudomonas. INTRODUÇÃOA rigor, define-se como xenobiótico qualquer substância química estranha ao sistema biológico humano originada externa ou internamente a ele.1 São utilizados na agricultura com finalidade de controle fitossanitário e aumento da produtividade. Muitos desses produtos têm efeito nocivo ao meio ambiente, devendo ser recomendados de maneira criteriosa a fim de reduzir o risco de impacto ambiental. 2A degradação microbiana é considerada o mais importante processo que determina a persistência de pesticidas no solo. Os micro--organismos, devido à sua capacidade degradadora, participam de forma significativa na eliminação ou redução acentuada dos níveis desses compostos empregados na agricultura. 3Bactérias do gênero Pseudomonas destacam-se neste sentido, sendo descritas como capazes de degradar os mais diversos compostos, 4-7 incluindo o paclobutrazol (PBZ). 8,9 O metabolismo microbiano pode resultar na completa mineralização das moléculas orgânicas, isto é, sua conversão para CO 2 , água, íons inorgânicos e energia.10 Em contrapartida, muitos micro--organismos são capazes de transformar parcialmente os compostos químicos em produtos sem produção de energia para o seu crescimento. Paclobutrazol is a plant growth retardant which is used world-wide for increasing the yield of cereal crops. However, this compound remains active in the soil for several years and can severely affect the growth and development of subsequent crops, mainly by reducing vegetative vigor. The aim of this work was to develop and validate methods for the determination of paclobutrazol concentrations by both high performance liquid chromatography and spectroscopy. Both methods were satisfactory and showed appropriately low quantification limits. INTRODUÇÃOPaclobutrazol é um regulador do crescimento de plantas usado no mundo inteiro, tanto para esta finalidade como para aumentar o rendimento de colheitas. Entretanto, é um composto que permanece ativo no solo por vários anos, podendo afetar o crescimento e desenvolvimento de colheitas subseqüentes, principalmente pela redução do vigor vegetativo do solo 1 . A necessidade de reduzir o impacto ambiental tem despertado o interesse científico para o estudo sobre a biodegradação de tais moléculas. A ação dos microrganismos do solo sobre estas substâncias constitui-se em um importante mecanismo para dissipação da molécula no ambiente 2 . Estudos em laboratório sobre a biodegradação de paclobutrazol, geralmente, são realizados em meios de cultura contendo este composto em concentrações variando entre 0,25 e 1 g/L 1,3 , uma vez que a quantidade utilizada no campo varia entre 0,25 e 1,5 g/L.A literatura apresenta alguns métodos para a determinação de paclobutrazol ([2RS,3RS]-1-(4-clorofenil)-4,4-dimetil-2-(1H-1,2,4-tiazolil-1)pentanol-3), que utilizam cromatografia gasosa 4,5 ou cromatografia líquida de alta eficiência (CLAE) acoplada à espectrometria de massa 6 , o que torna a metodologia muito cara. No presente trabalho foram desenvolvidos dois métodos analí-ticos, por CLAE e por espectroscopia UV, para a determinação de paclobutrazol em amostras aquosas, como por ex., aquelas provenientes de experimentos de biodegradação deste composto, bem como do produto comercial Cultar 250 SC, contendo aproximadamente 250 g/L de paclobutrazol. PARTE EXPERIMENTAL MaterialFoi utilizado o padrão de paclobutrazol (PBZ) da Sigma e o produto comercial na forma de uma suspensão de PBZ (Cultar 250 SC). O metanol foi de grau cromatográfico com 99,99% de pureza (Merck) e a água utilizada foi de grau ultrapuro (Merck, Easy Pure).
