Perairan muara sungai Banjir Kanal Barat merupakan salah satu perairan yang penting untuk menopang kehidupan organisme didalamnya, serta penting bagi ekonomi masyarakat kota Semarang. Tujuan penelitian ini adalah mengetahui pola sebaran klorofil-a dan fitoplankton, mengetahui hubungan klorofil-a, nitrat, dan fosfat dengan fitoplankton serta mengetahui tingkat saprobitas perairan sebagai indikator tingkat pencemaran. Penelitian dilaksanakan pada bulan April 2019 di muara sungai Banjir Kanal Barat, Semarang. Pengambilan sampel air dilakukan pada saat kondisi pasang dan surut dengan teknik purposive sampling. Pengambilan sampel fitoplankton dengan metode pasif. Analisis data dengan metode T test, regresi polinomal dan PCA. Analisis status perairan dengan indeks saprobitas dan tropik saprobik indeks. Hasil penelitian ini adalah konsentrasi klorofil-a saat pasang berkisar 0,016 – 2,394 mg/m3 sedangkan saat surut berkisar 0,102 – 1,051 mg/m3. Kelimpahan fitoplankton saat pasang yaitu 33 – 127 sel/l, sedangkan kelimpahan fitoplankton saat surut berkisar antara 18 – 200 sel/l. Sebaran horizontal klorofil-a dan fitoplankton pada saat pasang maupun surut lebih tinggi pada lokasi sungai dan muara dan berkurang pada lepas pantai. Hubungan yang kuat dari analisa regresi polinomial dan PCA yaitu pada fitoplankton dengan nitrat saat pasang serta klorofil-a dengan nitrat saat surut. Nilai SI pada saat pasang menunjukkan β-mesosaprobik dan saat surut oligosaprobik. Nilai TSI pada saat pasang dan surut menunjukkan oligosaprobik.
