Resumo -O objetivo deste trabalho foi avaliar a resistência parcial de genótipos de soja ao fungo Phakopsora pachyrhizi. Calcularam-se o número médio de pústulas, a severidade e a área abaixo da curva de progresso da doença. Foram encontradas diferenças significativas entre os genótipos quanto ao número médio de pústulas e severidade, aos 12 dias após a inoculação. A análise de agrupamento permitiu a discriminação de genótipos parcialmente resistentes. Epidemiological characters and the use of cluster analysis for characterizing partial resistance to soybean rustAbstract -The objective of this work was to evaluate the partial resistance of soybean genotypes against Phakopsora pachyrhizi. Resistance characteristics were: average number of pustules, rust severity and the area under the disease progress curve. Significant differences were found among the genotypes for the average number of pustules and rust severity. Multivariate analysis allowed the discrimination of partially resistant genotypes. O controle da doença tem exigido combinação de práticas culturais, a fim de que sejam minimizados os danos e as perdas. Entre os métodos de controle, o único disponível, no momento, é o químico, por meio de fungicidas. Estratégias de controle, como a utilização de cultivares com resistência parcial ao fungo, são desejáveis para o manejo eficiente da cultura.A resistência pode ser definida como a habilidade do hospedeiro em impedir o crescimento e o desenvolvimento do patógeno (Parlevliet, 1997). A resistência parcial tem como característica a redução da taxa da epidemia, pela diminuição do número e tamanho das lesões, pela diminuição da produção de uredinosporos, e pelo aumento do período latente. Isso faz com que a população do patógeno seja reduzida, e que seja diminuída a quantidade de inóculo e, conseqüentemente, a intensidade da doença (Wang & Hartman, 1992). Este tipo de resistência torna-se visível após a resistência não durável ou monogênica ter sido superada por uma nova raça do patógeno (Parlevliet, 1997). Segundo Parlevliet (1983), a seleção para resistência parcial na presença de genes maiores pode ser indesejável, uma vez que o efeito dos genes maiores pode suprimir o efeito dos genes menores sob determinadas condições experimentais. RESUMOOs objetivos deste trabalho foram determinar o estádio fenológico ideal de plantas de soja para inoculação de Sclerotinia sclerotiorum, bem como o período apropriado de contato do inóculo com o tecido da planta para detectar resistência parcial de cultivares de soja ao patógeno. Foram testadas duas cultivares de soja, MG/BR-46 (Conquista) e M-SOY 8200, com plantas em cinco estádios fenológicos diferentes (V 1 , V 2 , V 3 , V 4 e R 1 ) e a inoculação envolveu diferentes períodos de contato de discos de cultura de um isolado de S. sclerotiorum escolhido por sua elevada agressividade à soja. Observou-se que a severidade da doença decresceu em proporção direta à idade das plantas, determinando-se V 1 e V 2 (folhas e hastes) e V 3 (folhas) como estádios ideais de inoculação para a avaliação de resistência de cultivares de soja. O contato contínuo do inóculo com o tecido vegetal proporcionou bons resultados para a inoculação de S. sclerotiorum, tanto em órgãos destacados como em órgãos ligados às plantas. Foram avaliados 90 genótipos de soja em inoculações sobre folhas destacadas pela colocação de discos de cultura em contato permanente com os folíolos das plantas. Apenas 19 genótipos comportaram-se como resistentes ou moderadamente resistentes e, dentre estes, apenas dois foram moderadamente resistentes em inoculações nas plantas. ABSTRACT Evaluation of soybean resistance to Sclerotinia sclerotiorum at different phenological stages and over different periods of exposure to inoculumThe objectives of this work were to determine the most adequate phenological stage of soybean plants for inoculation of Sclerotinia sclerotiorum and the period of contact between inoculum and plant tissues to detect partial resistance of soybean to the pathogen. Plants of two soybean cultivars, MG/BR-46 (Conquista) and M-SOY 8200 were inoculated at five different phenological stages (V 1 , V 2 , V 3 , V 4 and R 1 ). Inoculation involved different periods of contact between culture disks of an isolate of S. sclerotiorum which was selected based on high aggressiveness against soybean. It was observed that disease severity decreased as plants became older and it was concluded that V 1 and V 2 (leaves and stems) and V 3 (leaves) were ideal stages for inoculation during tests aimed at determining levels of resistance in soybean cultivars. Continuous contact of inoculum with plant tissues yielded good results for inoculation of S. sclerotiorum, either onto detached organs or onto organs connected to living plants. Ninety genotypes of soybean were evaluated by placing culture disks in continuous contact with folioles. Only 19 genotypes were moderately resistant and, among these, only two were moderately resistant in inoculation involving whole plants. The molecular marker technology has been used on mapping of quantitative trait loci (QTLs) associated with plant resistance. The objectives of this research were to estimate genetic parameters and to map genomic regions involved in the resistance to gray leaf spot in maize. Ninety F 3 families from the BS03 (susceptible) and BS04 (resistant) cross were used. Field trials were performed using a 10 9 10 square lattice design with three replications. Data from 62 SSR markers were used for linkage analysis. The locations of the QTLs on the linkage groups were determined by composite interval mapping method and the phenotypic variance explained by each marker was determined by regression analysis. Several QTLs associated to disease resistance were identified in the population BS03 9 BS04. Some QTLs showed significant effects over the different environments studied. The existence of significant QTLs in common among different environments indicates these genomic regions as possible new tools for marker-assisted selection in maize breeding programs.
O presente trabalho teve como objetivo avaliar a reação quanto à resistência parcial causada pelo fungo Phakopsora pachyrhizi Sydow., em 68 genótipos de soja (Glycine max L. Merril). O experimento foi realizado em Uberlândia-MG, em casa de vegetação, durante o período de dezembro de 2004 a fevereiro de 2005 e constou de três épocas de avaliação. Foram avaliadas as seguintes características de resistência: Período Latente Médio (PLM), número médio de pústulas por folíolo e severidade da ferrugem. Com base nas variáveis, número médio de pústulas por folíolo e severidade da ferrugem calculou-se a área abaixo da curva de progresso da doença. Após, realizou-se a análise de variância, e as médias foram comparadas pelo teste de Scott Knott, ao nível de 5% de probabilidade. Foram encontradas diferenças significativas entre os genótipos para número médio de pústulas por folíolo e severidade da ferrugem, aos doze dias após a inoculação. O mesmo foi encontrado para área abaixo da curva de progresso da doença (AACPD) para severidade. Com base no período latente médio discriminaram-se dois grupos de genótipos: parcialmente resistentes e suscetíveis. Presume-se que a Resistência Parcial (RP) à ferrugem asiática da soja deve ser avaliada em condições de média epidemia, que aconteceu na segunda época de avaliação para todos os caracteres avaliados. O agrupamento de genótipos, pela análise multivariada de componentes principais, permitiu a discriminação de genótipos parcialmente resistentes. Três genótipos (números 4, 41, e 42), referentes aos parentais Cristalina e IAC 100, apresentaram maior resistência parcial á ferrugem da soja.
Biological control agents are alternatives to chemical pesticides in the management of plant diseases. Currently, hundreds of bioproducts are commercially available in international market varying mainly in antagonistic microorganisms and formulation. We screened four Trichoderma-based products as to their efficacy in controlling Sclerotinia stem rot (SSR) under protected and field environments and their effect on soybean seeds' sanity and physiological qualities. We also tested application technologies through seed microbiolization and foliar spraying to deliver the microorganisms, and their compatibility with chemical fungicides. In vitro assays showed that all Trichoderma strains were antagonistic to S. sclerotiorum evidencing hyperparasitic activity. Moreover, the bioproducts reduced fungi incidence on soybean seeds, promoted faster seedling emergence and did not hamper seeds' vigor. Increases of 14 and 37% were registered for root length and shoot fresh weight over that of the untreated control indicating potential application of the bioproducts as soybean growth promoters. Thiophanate-methyl and procymidone were the most compatible, without drastically affecting spore germination or mycelium growth. Under field conditions, all Trichoderma strains reduced SSR incidence and increased soybean grain yield. Formulation interferes on bioproducts' viability and efficacy deserving special attention upon development.
