Kvasny Prum. 64(4): 161-166 The Enterobacterales order contains 7 families with more than 40 bacterial genera which are isolated from various types of environment. The genera Shimwellia, Obesumbacterium, Rahnella, Citrobacter, Klebsiella, Raoultella, Serratia and Enterobacter are often found in the beer brewing process. The ability of these bacteria to grow and multiply in the finished beer is minimal. The bacteria occur primarily as contamination of yeast. They are harmful because these bacteria produce sensory undesirable substances (e.g. dimethylsulfide, diacetyl, acetoin) at the beginning of the main fermentation and get into the finished beer. Some species are involved in the formation of N-nitrosamines harmful for health. Here we present an overview of the basic morphological and physiological properties of these bacteria and describe their importance from the point of view of harmfulness in the beer brewing process.
This review focuses on autochthonous wine yeasts that occur naturally in a specific geographical area, and their role in wine production. The distribution of individual species changes during ripening and processing of grapes and fermentation of the grape must. The composition of autochthonous microflora is influenced by many factors including interspecific interactions. Autochthonous yeast populations lend to wine flavor and taste typical for a specific geographical area. The article describes the fermentation of grape must, changes of autochthonous yeast microflora during spontaneous or combined fermentation, and includes an overview of the basic species of autochthonous yeasts.
The growth of 7 strains belonging to the order of Enterobacterales, represented by the species of Citrobacter Freundii, Enterobacter cloacae, Escherichia coli, Klebsiella oxytoca, Obesumbacterium proteus, Rahnella aquatilis, Raoultella terrigena, Serratia marcescens and Shimwellia pseudoproteus, was monitored on selected cultivation media. Three types of agars - Endo, MacConkey and Chromocult Coliform agar together with two incubation temperatures of 28 and 37 °C were tested under aerobic conditions. The aim of the study was to detect such essential enterobacteria harmful to beer that cannot be proven at 37 °C, which is the temperature usually used in operational laboratories in breweries. Our results showed that most of the tested strains of enterobacteria were able to grow at 28 °C on all selected types of agar. The exception was just the representatives detection of which is problematic at 37 °C. Nevertheless, a little or no growth was always observed on just one of the tested media.

ČástKlíčová slova: adaptace, bakterie mléčného kvašení, kontaminace piva, kultivační metody, Lactobacillus, Pediococcus ■ 1 ÚVOD Tato studie navazuje na články týkající se problematiky kultivač-ních metod detekce bakterií mléčného kvašení: literární rešerši zahrnující jednak základní charakteristiku této skupiny bakterií z hlediska rizika pro výrobu piva a jednak stručný přehled vývoje kultivačních médií (Matoulková a Kubizniaková, 2015) a článek shrnující výsled-ky laboratorního testování vybraných půd určených pro detekci mléčných bakterií (Kubizniaková a Matoulková, 2016). Z úvodního článku vyplynulo, že nejčastěji používanými půdami pro detekci bakterií mléčného kvašení jsou různě modifi kované půdy MRS a NBB. V navazující studii byla jako nejlepší vyhodnocena půda MRS a její modifi kace, K-MRS (s aplikací enzymu katalasy) a B-MRS (s přídav-kem piva), na nichž narostly všechny bakteriální kmeny nejrychleji a poskytovaly největší kolonie (Kubizniaková a Matoulková, 2016). Ve studii byly použity čisté laboratorní kmeny, které sice byly izolovány z piva nebo z pivovarského prostředí, ale již dlouhou dobu byly uchovávány v laboratorních podmínkách, tj. bez hořkých chmelových látek, alkoholu, v médiu s optimálním složením. Takové kmeny pak nemusí růst např. na půdách obsahujících pivo, neboť během dlouhodobého skladování v laboratorních podmínkách ztratily schopnost růst v přítomnosti hořkých chmelových látek a alkoholu (Suzuki et al., 2006). V literatuře byl u některých mléčných bakterií popsán stav "obtížné kultivovatelnosti", který souvisí s jejich adaptací na specifi cké prostředí, v tomto případě na pivo. Tento stav, nazý-vaný také stavem "životaschopnosti ale nekultivovatelnosti" (tzv. Ve studii byl sledován růst souboru 10 kmenů bakterií rodu Lactobacillus a Pediococcus na ztužených kultivačních půdách, např. MRS agaru a jeho různých modifi kacích, na půdě Raka-Ray, NBB, UBA atd. Sledována byla rychlost tvorby bakteriálních kolonií a jejich velikost. Kmeny byly na kultivační média naočkovány přímo z piva, a to ve dvou variantách -s předchozí postupnou adaptací na pivo a bez fáze adaptace. Postupné převádění bakterií do půdy se stoupající koncentrací piva (fáze adaptace) u některých kmenů zvýšilo jejich schopnost dlouhodobého přežívání v pivu. Při následném testování kultivačních médií byla jako nejúčinnější vyhodnocena půda MRS a její modifi kace. Půda Raka-Ray byla vyhodnocena jako nejméně vhodná pro detekci bakterií mléčného kvašení.
