RESUMO: O uso do solo com cultivos anuais durante longo tempo, especialmente quando são utilizados manejos não conservacionistas, como arações e gradagens, normalmente são responsáveis pela degradação mais intensa de suas propriedades. Este trabalho avaliou as propriedades do solo em três áreas sob cultivo agrícola e uma área nativa de Cerrado, verificando as alterações ocorridas nos atributos físico-químicos do solo após dois, cinco e oito anos sob plantio de soja. Este trabalho tem como objetivo avaliar a quebra da dormência da semente de girassol em diferentes métodos. O conhecimento acerca dos aspectos germinativos de sementes silvestres é de extrema importância para a manutenção e propagação dos recursos genéticos dos bancos de germoplasma. No presente trabalho foi utilizado 3 tratamentos com 4 repetições de 50 semente em cada repetição, no total de 600 sementes utilizadas: No primeiro tratamento foi utilizado ácido giberélico de 1000 mg-l, no segundo tratamento, escarificação mecânica manual, no terceiro e último tratamento foi a testemunha sem nenhum método para quebra. O tratamento com ácido giberélico foi o que mostrou melhores resultados na quebra de dormência das sementes de girassol.
The objective of the current study was to measure the genetic variability of natural populations of Hancornia speciosa using RAPD type molecular markers to assay variation in existing sampled genotypes, using morphological variables, and so assess germplasm bank composition. Morphological and chemical characteristics H. speciosa fruits and seeds were evaluated using descriptive statistics and principal components analysis. Cluster analyzes was conducted using Jaccard's similarity index, via the UPGMA hierarchical agglomerative method. Phenotypic variability was found in the two studied populations. However, variability was higher in the São Judas population, where the variables: pulp yield and soluble solids content were higher than those in the Canaã population. High genetic variability was found in both study populations, and between- and within-population morphological and genetic variation was present in the studied populations. The nine primers generated 70 bands, of which 68 were polymorphic, with the primers A-08 and C-04 generating the highest number of polymorphic bands. The two populations differ principally in the pulp ratio and the proportion of total solids in the pulp (°Brix). RAPD markers used gave acceptable results and, to initiate the Federal University of Tocantins Active Mangaba Germplasm Bank, seven genotypes were sampled from the Canaã population and five from the São Judas Tadeu population.
The efficient use of genetic resources-stored in germplasm collections can be maximized if morphoagronomic and molecular information on the accessions is made available. To achieve this, a collection that is well-structured, well-curated and easily accessible (the core collection) is required. Consequently, the objective of the current study was to characterize 80 landrace accessions from the maize core collection of the Federal University of Viçosa (UFV), and assay thenngenetic diversity of the various landraces, considering grain type and ecogeographic origin. For this, AFLP analysis was performed using 12 primer combinations. Genetic diversity of the collection was quantified with the UPGMA method, using the Jaccard Index to quantify dissimilarity. The core collection was divided into four sub-populations by grain type, and into six sub-populations based on ecogeographic origin. Genetic diversity analysis was performed both within and between sub-populations. A high level of genetic variability was found among the landrace accessions of UFV Core Collection, principally among those accessions with dentate type grains.Classification by grain type and ecogeographic origin allowed genetically divergent groups to be distinguished.Resumo: A utilização eficiente de recursos genéticos, armazenados em coleções de germoplasma, pode ser maximizada se informações morfo-agronômicas e moleculares sobre os acessos forem disponibilizadas. Para isso, é necessária a existência de uma coleção racionalizada, refinada e estruturada, chamada de coleção núcleo. Assim, objetivou-se com este trabalho caracterizar molecularmente 80 acessos landraces da coleção núcleo de milho da Universidade Federal de Viçosa (UFV) e verificar a diversidade genética com relação à estratificação, considerando tipo de grão e origem ecogeográfica. Para isso, foi realizada análise de AFLP, utilizando-se 12 combinações de primers. A diversidade genética da coleção foi quantificada utilizando o método de UPGMA, tendo como medida de dissimilaridade o complemento do índice de Jaccard. A coleção núcleo foi estratificada em quatro sub-populações quanto ao tipo de grãos e seis quanto à origem ecogeográfica. Foi realizada análise de diversidade genética tanto entre quanto dentro das subpopulações. Foi verificada grande variabilidade genética entre os acessos landraces da Coleção Núcleo da UFV, principalmente entre os acessos com grãos do tipo dentado, e que a estratificação quanto ao tipo de grãos e origem ecogeográfica foi eficiente para a obtenção de grupos geneticamente divergentes.
