RESUMO -Para determinação da dose inseminante ideal na fertilização artificial de ovócitos de jundiá cinza (Rhamdia quelen), foram selecionadas duas fêmeas com papila urogenital avermelhada, abdômen arredondado e liberação de ovócitos de cor e tamanho uniforme e quatro machos que apresentavam liberação de sêmen sob pressão da papila genital. As fêmeas e os machos receberam duas doses de extrato de hipófise de carpa (doses inicial e final de 0,5 e 0,25 mg.kg -1 e de 5,0 e 2,5 mg.kg -1 , para fêmeas e machos, respectivamente). As duas fêmeas produziram 169 g de ovos contendo 1.141 ovócitos.g de ovócitos -1 e os machos, um total de 23,6 mL de sêmen. Do sêmen dos quatro machos foi realizado um pool e analisadas as seguintes características: concentração espermática (1,97x10 10 espermatozóides.mL -1 ), índice de sobrevivência espermática (96%) e morfologia espermática (66,9% de espermatozóides normais). No ensaio de fertilização, utilizou-se delineamento experimental inteiramente casualisado, composto de cinco tratamentos (6x10 3 , 6x10 4 , 6x10 5 , 6x10 6 , 2x10 7 espermatozóides.ovócito -1 ) e quatro repetições. Após 8 horas de incubação, foram analisadas as taxas de fertilização para cada tratamento. A melhor relação espermatozóide.ovócito -1 foi de 89.497 espermatozóide.ovócito -1 , o que provocou taxa de fertilização máxima teórica de 86,68%. After eight hours of incubation, the fertilization rates for each treatment were analyzed. The best spermatozoa.oocyte -1 rate was of 89.497 spermatozoa.oocyte -1 that produced a maximum fertilization rate of 86.68%.

RESUMO -Um experimento foi conduzido com o objetivo de avaliar os desempenhos reprodutivo e zootécnico e a deposição de lipídios no tecido hepático de fêmeas de tilápia-do-nilo alimentadas com rações contendo diferentes níveis de energia digestível, obtidos pela inclusão de óleo de soja. Foram utilizados 100 reprodutores e 300 reprodutoras, distribuídos em delineamento inteiramente casualizado com cinco tratamentos e quatro repetições. Os reprodutores, machos e fêmeas, foram alimentados com rações contendo, por quilograma, 35% de proteína bruta e 2.700, 2.950, 3.200, 3.450 ou 3.700 kcal de energia digestível. Os animais foram mantidos em hapas por 93 dias. O desempenho reprodutivo e zootécnico não foi influenciado pelos níveis energéticos das rações, o que pode estar relacionado ao curto período de tempo de alimentação com as dietas experimentais. Contudo, o aumento nos níveis de energia teve efeito linear nas deposições lipídicas nos hepatócitos.Desta forma, os níveis crescentes de energia digestível nas rações fornecidas às fêmeas de tilápia-do-nilo, obtidos pela inclusão de óleo de soja, apenas aumentam o tempo de sobrevivência das larvas ao jejum e a deposição de lipídios nos hepatócitos das fêmeas. Palavras-chave: fígado, nutrição, Oreochromis niloticus, ovários, peixe, reprodução Reproductive and animal performance and lipids deposition in hepatocytes for Nile tilapia females fed different levels of energy in the rationABSTRACT -The experiment was carried out to evaluate the reproductive and animal performance and lipids deposition in the liver tissue of the Nile tilapia female, fed diets containing different levels of digestible energy, obtained by soybean oil inclusion. Three hundred breeding females and 100 breeding males were used, distributed in a completely randomized design with five treatments and four replications. The broods were fed diets containing 35% crude protein and 2,700; 2,950; 3,200;3,450 and 3,700 kcal energy digestible/kg feed. The animals were stocked in hapas for 93 days. The reproductive and animal performance were not influenced by the energy levels. These results may be associated with the short period of time that the brood were fed the experimental diets. However, increasing energy levels caused a linear increase in lipids inclusions in hepatocytes. Thus, the increasing levels of digestible energy provided to Nile tilapia females, obtained by including soybean oil, only promoted increased lipid deposition in hepatocytes.

Summary The objective of this work was to evaluate the sperm motility of 13 Steindachneridion parahybae males using open‐source software (ImageJ/CASA plugin). The sperm activation procedure and image capture were initiated after semen collection. Four experimental phases were defined from the videos captured of each male as follows: (i) standardization of a dialogue box generated by the CASA plugin within ImageJ; (ii) frame numbers used to perform the analysis; (iii) post‐activation motility between 10 and 20 s with analysis at each 1 s; and (iv) post‐activation motility between 10 and 50 s with analysis at each 10 s. The settings used in the CASA dialogue box were satisfactory, and the results were consistent. These analyses should be performed using 50 frames immediately after sperm activation because spermatozoa quickly lose their vigor. At 10 s post‐activation, 89.1% motile sperm was observed with 107.2 μm s−1 curvilinear velocity, 83.6 μm s−1 average path velocity, 77.1 μm s−1 straight line velocity; 91.6% were of straightness and 77.1% of wobble. The CASA plugin within ImageJ can be applied in sperm analysis of the study species by using the established settings.
