Genetic diversity and relationships within and among nine species of Coffea, one species of Psilanthus and the Piatã hybrid from the Coffee Germplasm Collection of Instituto Agronô-mico de Campinas (IAC), Brazil were assessed using RAPD markers. Genetic diversity and relationships were evaluated by proportion of polymorphic loci (P), Shannon's genetic index (H 0 and G 0 ST ) and clustering analysis. The overall RAPD variation among all accessions was mostly partitioned between rather than within species. However, C. canephora and C. liberica showed a high genetic diversity within the species (H 0 sp = 0.414 and H 0 sp = 0.380, respectively) and this was highly structured (high G 0 ST ). Genetic diversity from C. congensis and C. arabica was also structured, but with lower levels of genetic diversity (H 0 sp = 0.218 and H 0 sp = 0.126, respectively). The results were consistent with agronomic and molecular studies and demonstrated that the IAC Coffea Collection is representative of the phylogenetic structure observed in the genera. This study devises sampling strategies for coffee germplasm collections and provides genetic diversity parameters for future comparisons among them.
-The objective of this work was to identify genes that could be used as suitable markers for molecular recognition of phenological stages during coffee (Coffea arabica) fruit development. Four cultivars were evaluated as to their differential expression of genes associated to fruit development and maturation processes. Gene expression was characterized by both semi-quantitative and quantitative RT-PCR, in fruit harvested at seven different developmental stages, during three different seasons. No size polymorphisms or differential expression were observed among the cultivars for the evaluated genes; however, distinct expression profiles along fruit development were determined for each gene. Four out of the 28 evaluated genes exhibited a regular expression profile in all cultivars and harvest seasons, and, therefore, they were validated as candidate phenological markers of coffee fruit. The gene α-galactosidase can be used as a marker of green stage, caffeine synthase as a marker of transition to green and yellowish-green stages, and isocitrate lyase and ethylene receptor 3 as markers of late maturation.Index terms: Coffea arabica, cup quality, differential expression, fruit phenology, ripening, qRT-PCR. Perfil da expressão gênica durante o desenvolvimento de frutos de café e identificação de genes marcadores para fases fenológicasResumo -O objetivo deste trabalho foi identificar genes que possam ser utilizados como marcadores moleculares para reconhecimento de fases fenológicas, durante o desenvolvimento de frutos do cafeeiro (Coffea arabica). Quatro cultivares foram avaliadas quanto à expressão diferencial de genes associados a processos de desenvolvimento e maturação de frutos. A caracterização da expressão gênica foi realizada pelas técnicas de RT-PCR semi-quantitativa e quantitativa, em frutos coletados em sete estádios de desenvolvimento, durante três safras. Não foi observado nenhum polimorfismo de tamanho ou expressão diferencial entre as cultivares, para os genes avaliados; porém, perfis de expressão distintos durante o desenvolvimento dos frutos foram determinados para cada gene. Quatro entre os 28 genes avaliados apresentaram perfil de expressão constante, em todas as cultivares e safras e, portanto, foram validados como genes candidatos a marcadores fenológicos de frutos de cafeeiro. O rendimento intrínseco do café, relação percentual entre a massa de dois grãos normais tipo chato e do respectivo fruto que os contém, foi estudado em seis grupos de germoplasma de Coffea, com o objetivo de se conhecer a variabilidade genética para essa característica. Investigou-se o rendimento intrínseco de Coffea arabica em um grupo de cinco cultivares de porte baixo, em outro contendo 22 cultivares e seleções e, ainda, em outro grupo com 79 cultivares, variedades e formas botânicas, mutantes e acessos da Etiópia. Em C. canephora, foram analisados três acessos da variedade kouilou e 10 acessos da variedade robusta. Investigaram-se ainda, outras oito espécies do gênero Coffea. Observou-se considerável variabilidade genética tanto entre representantes de C. arabica quanto de C. canephora, assim como entre as diferentes espécies do gênero Coffea. A amplitude de variação nos valores de rendimento intrínseco referente ao último grupo foi bem maior que a de qualquer outro grupo estudado. A magnitude das variações observadas e as implicações econômicas do rendimento intrínseco indicam que essa característica pode ser utilizada como um critério adicional de seleção no melhoramento de C. arabica e C. canephora.
O objetivo deste trabalho foi avaliar o efeito do ano agrícola sobre os atributos fenológicos e agronômicos em diferentes cultivares de cafeeiro arábica. Os atributos fenológicos determinados foram estádios de frutificação, de gema dormente até fruto seco e duração do ciclo. Como atributos agronômicos, foram avaliados o rendimento, a produtividade, os tipos e o tamanho de grãos. O período entre antese até fase chumbinho não variou entre as cultivares para o ano agrícola 2004/2005, já para o ano agrícola seguinte a cultivar 'Icatu Precoce' apresentou antecipação da fase chumbinho, e a cultivar 'Obatã' atrasou o início do ciclo fenológico em relação às demais cultivares estudadas. A duração do ciclo variou com o ano agrícola, o que possibilitou a confirmação da influência dos efeitos ambientais nessa característica. Os atributos agronômicos (produtividade, rendimento, tipo e tamanho de grãos), independentemente das cultivares, também variaram com o ano agrícola.
