RESUMOA conservação das sementes de araucária é amplamente comprometida em função das suas características recalcitrante, o que dificulta o planejamento de ações de recuperação das populações degradadas. Desta forma, objetivou-se com o trabalho monitorar as alterações fisiológicas em sementes de araucária, sob diferentes condições de armazenamento, visando obter subsídios para a conservação de sua viabilidade e vigor. Avaliou-se a qualidade fisiológica das sementes recém-colhidas e a cada 60 dias, ao longo do armazenamento em ambiente de laboratório sem controle térmico, em refrigerador (5 o C) e em freezer (-18 o C), até o período de 180 dias. Após cada período amostral, avaliou-se a viabilidade (teste de germinação e tetrazólio) e o vigor das sementes (envelhecimento artificial, índice de velocidade de germinação -IVG e condutividade elétrica). Observou-se redução na porcentagem de plântulas normais ao longo do período de armazenamento das sementes de araucária. A conservação em freezer e em ausência de controle térmico causou a perda total de viabilidade das sementes aos 60 e 180 dias de armazenamento, respectivamente. Já o armazenamento em refrigerador favoreceu a conservação da viabilidade das sementes, com 64% de germinação aos 180 dias de armazenamento, um evento associado à redução da atividade metabólica das sementes. Com base nos testes de viabilidade e vigor, conclui-se que o armazenamento em refrigerador proporcionou a conservação das sementes de araucária por tempo superior em comparação às demais condições de armazenamento. -This research was aimed at studying effects of storage and accelerated aging on germination and profile of storage proteins in Handroanthus albus seeds. These were stored into a cold chamber (± 8 o C; RH ± 40%) and after periods of 0, 3, 6, 9, and 12 months of storage, were subjected to accelerated aging for 0, 24, 48, 72, and 96 hours. Relationships between germination and proteins profile were assessed. Germination test was performed at 25 °C, under constant light. For protein extraction, 125 mg of seeds were macerated in 2 mL of extraction buffer (1M TrisHCl; pH 8.8) and applied to SDS-PAGE polyacrylamide gel at 80 V .15 h -1 . Twelve month storage, combined with 72 hours accelerated aging have increased germination in approximately 65% when compared to non-aged seeds or to seeds with 24 h of accelerated aging. Besides beneficial effects, degradation and synthesis of different proteins were observed. It was concluded that germination of Handroanthus albus seeds, when not subjected to accelerated aging, is favored by storage in cold chamber during three to six months, or from nine to 12 months when subjected to accelerated aging process. Storage proteins may be associated to those increases, and hence further studies are needed.Index terms: yellow "ipê", protein, Tabebuia alba. As sementes foram armazenadas em câmara fria e ao 0, 3, 6, 9 e 12 meses submeteu-se ao envelhecimento acelerado por 0, 24, 48, 72 e 96 h, e avaliou-se a germinação e o perfil proteico. O teste de germinação foi realizado sob luz constante a 25 °C. Para a extração das proteínas, utilizaram-se 125 mg de sementes maceradas com 2 mL do tampão de extração (1M tris HCl pH 8,8) e aplicadas em gel SDS-PAGE (3% e 15%) a 80 V .15 h -1 . O armazenamento por 12 meses associado a 72 h de envelhecimento acelerado aumentaram a germinação em aproximadamente 65% em relação às sementes não envelhecidas e envelhecidas por 24 h. Associado a este efeito benéfico, observouse a degradação e síntese de diferentes proteínas. Conclui-se que a germinação de sementes de Handroanthus albus não envelhecidas artificialmente é favorecida pelo armazenamento em câmara fria por três ou seis meses, e por nove ou 12 meses, associado ao envelhecimento acelerado. As proteínas de reserva podem estar associadas a este acréscimo e necessitam ser mais bem estudadas.Termos para indexação: ipê-amarelo, proteínas, Tabebuia alba.
Abstract:Reactive oxygen species (ROS) are present in all aerobic organisms, but the seed deterioration processes can increase its production. Oxidative stress caused by higher endogenous ROS may cause irreparable damage to cells, leading to the loss of seed viability. Considering that the enzyme superoxide dismutase (SOD) composes the first enzymatic antioxidant defense mechanism, this study aimed to evaluate zygotic embryos of Araucaria angustifolia recalcitrant seeds during storage regarding changes in SOD activity. Besides, some of the major alterations resulting from oxidative stress, e.g., lipid peroxidation and changes in proteins and DNA integrity were also measured. Approximately, 7 000 seeds were harvested from a population in Southern Brazil and stored for 180 days under laboratory (L, laboratory temperature), refrigeration (R, 5 ± 1 °C) and freezing (F, -18 ± 1 °C) conditions. The analysis of lipid peroxidation through TBARS levels, SOD activity, protein profile by electrophoretic separation, and integrity of genomic DNA were performed at 0, 60, 120, and 180 days of storage. The results revealed an increase in lipid peroxidation and SOD activity, especially during the L storage, a condition in which there was an extensive degradation of proteins. Some proteins (i.e., 45, 32, and 31 kDa) were expressed only in embryos stored under R and F conditions. No damage was observed in the nuclear DNA integrity for the evaluated period of seed storage at R and F conditions. The F samples maintained the biochemical traits of interest throughout the storage period, with the exception of the protein profile. However, such changes are limiting for the maintenance of seed viability. In conclusion, R storage can be indicated to delay the metabolic feature alterations that occur when seeds are exposed to conditions of natural ambient after harvest, extending the conservation period of A. angustifolia seeds. <p>Este trabalho objetivou avaliar a eficiência de tratamentos pré-germinativos na promoção da germinação e crescimento inicial de plântulas de palmiteiro. Os tratamentos avaliados foram: controle, imersão em solução de GA<sub>3</sub>, exposição ao etileno, imersão em água, imersão em H<sub>2</sub>SO<sub>4</sub>, escarificação mecânica, estratificação por 30 dias a 10°C, e escarificação seguida de estratificação. A imersão de sementes em solução de ácido giberélico (GA<sub>3</sub>; 2000 µL L<sup>-1</sup> durante 24 h) ou sua exposição ao etileno (1000 µL L<sup>-1</sup> durante 24 h) são eficientes para a promoção da emergência e crescimento inicial de plântulas de palmiteiro.</p>
