We report a combined experimental and computational investigation to understand the nature of the interactions between cobalt redox mediators and TiO(2) surfaces sensitized by ruthenium and organic dyes, and their impact on the performance of the corresponding dye-sensitized solar cells (DSSCs). We focus on different ruthenium dyes and fully organic dyes, to understand the dramatic loss of efficiency observed for the prototype Ru(II) N719 dye in conjunction with cobalt electrolytes. Both N719- and Z907-based DSSCs showed an increased lifetime in iodine-based electrolyte compared to the cobalt-based redox shuttle, while the organic D21L6 and D25L6 dyes, endowed with long alkoxy chains, show no significant change in the electron lifetime regardless of employed electrolyte and deliver a high photovoltaic efficiency of 6.5% with a cobalt electrolyte. Ab initio molecular dynamics simulations show the formation of a complex between the cobalt electrolyte and the surface-adsorbed ruthenium dye, which brings the [Co(bpy)(3)](3+) species into contact with the TiO(2) surface. This translates into a high probability of intercepting TiO(2)-injected electrons by the oxidized [Co(bpy)(3)](3+) species, lying close to the N719-sensitized TiO(2) surface. Investigation of the dye regeneration mechanism by the cobalt electrolyte in the Marcus theory framework led to substantially different reorganization energies for the high-spin (HS) and low-spin (LS) reaction pathways. Our calculated reorganization energies for the LS pathways are in excellent agreement with recent data for a series of cobalt complexes, lending support to the proposed regeneration pathway. Finally, we systematically investigate a series of Co(II)/Co(III) complexes to gauge the impact of ligand substitution and of metal coordination (tris-bidentate vs bis-tridentate) on the HS/LS energy difference and reorganization energies. Our results allow us to trace structure/property relations required for further development of cobalt electrolytes for DSSCs.

A simple change of the substituents in the bridging ligand allows tuning of the ordering temperatures, Tc, in the new family of layered chiral magnets A[M(II)M(III)(X2An)3]·G (A = [(H3O)(phz)3](+) (phz = phenazine) or NBu4(+); X2An(2-) = C6O4X2(2-) = 2,5-dihydroxy-1,4-benzoquinone derivative dianion, with M(III) = Cr, Fe; M(II) = Mn, Fe, Co, etc.; X = Cl, Br, I, H; G = water or acetone). Depending on the nature of X, an increase in Tc from ca. 5.5 to 6.3, 8.2, and 11.0 K (for X = Cl, Br, I, and H, respectively) is observed in the MnCr derivative. Furthermore, the presence of the chiral cation [(H3O)(phz)3](+), formed by the association of a hydronium ion with three phenazine molecules, leads to a chiral structure where the Δ-[(H3O)(phz)3](+) cations are always located below the Δ-[Cr(Cl2An)3](3-) centers, leading to a very unusual localization of both kinds of metals (Cr and Mn) and to an eclipsed disposition of the layers. This eclipsed disposition generates hexagonal channels with a void volume of ca. 20% where guest molecules (acetone and water) can be reversibly absorbed. Here we present the structural and magnetic characterization of this new family of anilato-based molecular magnets.

Our aim was to elucidate the physiological role of calpains (CAPN) in mammary gland involution. Both CAPN-1 and -2 were induced after weaning and its activity increased in isolated mitochondria and lysosomes. CAPN activation within the mitochondria could trigger the release of cytochrome c and other pro-apoptotic factors, whereas in lysosomes it might be essential for tissue remodeling by releasing cathepsins into the cytosol. Immunohistochemical analysis localized CAPNs mainly at the luminal side of alveoli. During weaning, CAPNs translocate to the lysosomes processing membrane proteins. To identify these substrates, lysosomal fractions were treated with recombinant CAPN and cleaved products were identified by 2D-DIGE. The subunit b 2 of the v-type H þ ATPase is proteolyzed and so is the lysosomal-associated membrane protein 2a (LAMP2a). Both proteins are also cleaved in vivo. Furthermore, LAMP2a cleavage was confirmed in vitro by addition of CAPNs to isolated lysosomes and several CAPN inhibitors prevented it. Finally, in vivo inhibition of CAPN1 in 72-h-weaned mice decreased LAMP2a cleavage. Indeed, calpeptin-treated mice showed a substantial delay in tissue remodeling and involution of the mammary gland. These results suggest that CAPNs are responsible for mitochondrial and lysosomal membrane permeabilization, supporting the idea that lysosomal-mediated cell death is a new hallmark of mammary gland involution.
