A New Synthesis of Tetrahydrocapillsrine, 0-Methylglornelliue and OospolsetonePyridine catalysed acylation of a number of homophthalic acids with various acid anhydrides has been extensively studied, leading to the synthesis of several substituted isocoumarins including the natural products mentioned above.Die Erkenntnis, dal3 Homophthalsauren oder deren Anhydride in Gegenwart von Pyridin rnit Saurechloriden]) oder Anhydriden2.3) acyliert werden kdnnen und entweder das eigentliche 4-Acylisochroman-l,3-diOn oder 3-Alkylisocumarin-4-carbons~uren bilden konnen, regte uns an, eine neue Methode zur Synthese von 3-Alkylisocumarinen zu entwickeln. Diese temperaturabhangige Reaktion liefert bei Raumtemperatur das gewiinschte Produkt (die Dione); bei hdherer Temperatur (100 "C) wird die Bildung von intramolekularen Umwandlungsprodukten bevorzugt. Diese Umwandlung von 4-Acylisochrornan-I ,3-dion mit 80proz. Schwefelsiiure 3) zu 3-Alkylisocumarin-4-carbonsauren bzw. zu temperaturabhangig decarboxylierten Produkten ist bekannt. Trotz einiger erfolgreicher Untersuchungen iiber solche Acylierungsreaktionen erfordert die wachsende Zahl natiirlich vorkommender 3-substituierter Isocumarine mit langen Seitenketten eine weitere Erprobung dieser Reaktion mit Anhydriden hdherer aliphatischer Sauren.In dieser Arbeit wird iiber die Kondensation von Homophthalsauren mit verschiedenen SBureanhydriden berichtet. Diese Studie ftihrte unter anderem zur neuen Synthese von Naturstoffen wie Tetrahydrocapillarin (7, 0-Methylglomellin (8) Methode C beruht darauf, dal3 die Acylierung substituierter Homophthalsauren bei 100 "C meistens unter gleichzeitiger Decarboxylierung zu 3,4-disubstituierten Ismumarinen ablauft. Nach dieser Methode entstand aus 3-Methoxy-a-methylhomophthal~~re~) und Acetanhydrid 0-Methyloospolacton (14), welches zu Oospolacton (15) lo, 11) entmethyliert wurde.Nach den genannten Methoden A -C wurden mehrere in Tab. 1 zusammengestellte Isocumarine synthetisiert.
