o objetivo de adequar a metodologia do teste de tetrazólio para sementes de Melanoxylon brauna. As sementes foram pré-condicionadas por imersão em água destilada a 25ºC, por 14 horas. O tegumento foi retirado e os embriões colocados submersos em solução de tetrazólio (pH 6,5) nas concentrações de 0,05, 0,1 e 0,3%, sendo mantidos no escuro à temperatura de 25ºC por 10 e 24 horas. Os embriões foram avaliados quanto à intensidade da coloração, presença de áreas leitosas, aspectos dos tecidos e localização da coloração. Os embriões foram classificados individualmente em 9 classes de viabilidade. As sementes também foram submetidas aos testes de germinação e emergência de plântulas. Os tratamentos que mais se aproximaram do teste padrão de germinação de sementes de M. brauna foi a concentração de 0,3 por 10 horas. A viabilidade é representada pelas classes 1, 2 e 3, caracterizadas pela coloração rósea do embrião.Palavras-chave: Germinação, sementes florestais, embrião. ADJUSTMENT OF TETRAZOLIUM METHODOLOGY FOR THE EVALUATION OF Melanoxylon brauna SEEDS VIABILITY INTRODUÇÃOA utilização de sementes de alta qualidade são fatores básicos para o sucesso na produção de mudas e implantação de povoamentos florestais com espécies nativas. Para isso, o sistema de controle de qualidade na indústria de sementes deve ser ágil, versátil e confiável, fornecendo resultados precisos e rápidos, com resultados que avaliem a qualidade fisiológica das sementes por períodos mais curtos (FRANÇA NETO et al., 1998).O teste do tetrazólio tem por objetivo determinar rapidamente a viabilidade das sementes, particularmente daquelas que possuem germinação lenta ou que não germinam após o teste de germinação por estarem dormentes (BRASIL, 1992). Esse teste tem assumido posição de destaque para algumas culturas, em razão do grande número de informações fornecidas, e por ser uma técnica rápida e pouco dispendiosa (KRYZANOWSKI et al., 1999).Sabe-se que o tempo de incubação, a concentração da solução e a avaliação adequada do teste são fundamentais para que se obtenham resultados confiáveis da viabilidade e vigor (OLIVEIRA et al., 2005). A metodologia do teste vem sendo aprimorada, principalmente para sementes cultivadas (KRYZANOWSKI et al., 1999), mas, para espécies florestais, os estudos e a padronização do método ainda são poucos, embora tenha havido aumento nas publicações nos anos mais recentes, com aplicações diversas. Assim, Mendes et al. (2009) padronizaram o teste para Parkia velutina na concentração de 0,5%,
