Depression is a common disease affecting more than 300 million people worldwide. Since has shown central nervous system effects in previous works, we aimed to investigate the effect of essential oil and its major compound, thymol on a corticosterone-induced depression model in mice. Male mice (20⁻25 g) received corticosterone (20 mg/kg, subcutaneously), once a day for 22 days. From the 16th day on, mice were grouped to receive either corticosterone or essential oil (100 and 200 mg/kg), or thymol (25 and 50 mg/kg) or fluoxetine (35 mg/kg) by gavage. The forced swimming test, tail suspension, open field, elevated plus maze and sucrose preference tests were performed from the 19th to 22nd day. Data were analyzed by ANOVA followed by the Student-Newman-Keuls as a post hoc test and the results were considered significant when < 0.05. It was shown that essential oil, thymol and fluoxetine decreased the immobility time in the tail suspension and forced swimming tests and none of these altered locomotor activity in the open field test. However, the drugs increased the amount of grooming. In the elevated plus maze, all drugs increased the number of entries and the time of permanence in the open arms. In the sucrose preference test, the essential oil, thymol and fluoxetine reversed anhedonia. These results suggest that the thymol and essential oil have an antidepressant-like effect, similar to fluoxetine. However, future studies should be encouraged to enhance understanding of the effects of essential oil and thymol for the treatment of depression.
Cerca de 20 % da população brasileira é afetada por transtornos de ansiedade, sendo mais prevalente entre mulheres, com idade entre 34 e 59 anos e o alprazolam é o segundo ansiolítico mais utilizado para tratar essa patologia. Quanto às características dos comprimidos, estes devem possuir estabilidades física e química, desintegrar-se no tempo esperado, ser pouco friáveis, além de apresentarsuperfície lisa e brilhante. O objetivo deste trabalho foi avaliar a qualidade de comprimidos de alprazolam 0,5 mg, efetuando a comparação entre medicamentos de referência, genérico e similar disponíveis comercialmente, quanto ao aspecto visual e aos ensaios de peso médio, dureza e friabilidade. Foram utilizados 270 comprimidos não revestidos de alprazolam 0,5 mg, provenientes de três laboratórios diferentes, sendo que 90 destes eram comprimidos de referência (grupo A), 90 genéricos (grupo B) e 90 similares (grupo C). Cada grupo era integrado por três subgrupos, de lotes diferentes, e cada subgrupo continha 30 comprimidos do mesmo lote, sendo em triplicata os ensaios. Assim, o grupo A foi constituído pelos lotes A1, A2 e A3; o grupo B foi integrado pelos lotes B1, B2 e B3 e o grupo C foi composto pelos lotes C1, C2 e C3. Todos os lotes foram aprovados nos testes de peso médio e friabilidade. Entretanto, na análise do aspecto visual, os medicamentos do grupo C presentaram lote e data de validade ilegíveis e no teste de dureza, dois comprimidos do lote C1 apresentaram dureza abaixo das especificações descritas pela Farmacopeia Brasileira 5ª edição.
<p>A avaliação da capacidade inibitória dos conservantes presentes nas amostras é fundamental para uma avaliação microbiológica eficaz. A inibição do crescimento de micro-organismos devido ao efeito bacteriostático ou fungistático da formulação pode ocasionar um resultado falso-negativo na análise microbiológica. A inibição do efeito conservante da formulação foi realizada através de dois métodos: a adição de polissorbato 80 a 0,4% e a diluição. A amostra foi preparada utilizando 10 g de cada amostra em 90 ml de salina a 0,9%, resultando em uma diluição 10<sup>-1</sup>. Foi adicionado também polissorbato 80 a 0,4% na solução salina com objetivo de neutralizar os parabenos e a imidazolidinil uréia nas formulações. Os ensaios foram realizados com objetivo de comprovar a ausência do efeito inibitório no crescimento de micro-organismos decorrente da atividade inibitória desses componentes. Foi observado o crescimento dos seguintes micro-organismos: <em>Staphylococcus aureus</em>, <em>Salmonella typhimurium</em> e <em>Pseudomonas aeruginosa</em> em todas as amostras e no controle positivo, significando a inativação do efeito inibitório das substâncias conservantes. A neutralização do efeito inibitório foi realizada através do uso do polissorbato 80 a 0,4% e da diluição 1:10.</p>
Cough is a common pediatric complaint, resulting in frequent doctor's office or hospital visits. In Brazil, the leaves of chambá (Justicia pectoralis) have been used extensively in herbal preparations for the treatment of many respiratory problems such as cough, bronchitis, and asthma. Thus, this study aimed to assess the effectiveness of chambá syrup for the treatment of cough and respiratory symptoms in children. Between November 2017 and February 2018, patients aged 1 to 12 years with respiratory symptoms and cough, and onset of symptoms greater than 48 h were enrolled in this trial. Participants were randomized to receive either chambá syrup in three daily doses or placebo. Effectiveness was assessed through a questionnaire, to measure the intensity of cough and respiratory symptoms, before the intervention and 48 h after, using Likert scale responses. A total of 114 children, 56 in the chambá group and 58 in the placebo group, participated in the study. After 48 h, there was a significant reduction in frequency of cough, severity of cough, severity of nasal congestion, and in improvement in the ability to sleep for both the child and caregiver in the chambá group (p< .0001 for all variables); for participants in the placebo group, there were no significant differences after intervention, in comparison with baseline. In this study population, chambá syrup was effective in the treatment of cough and respiratory symptoms, providing an overall improvement of symptoms when compared to placebo, as well as improving the ability to sleep.
