This study proposes a new approach to selecting a biofilm carrier for immobilization using dissolved oxygen (DO) microsensors to measure the thickness of aerobic and anaerobic layers in biofilm. The biofilm carriers tested were polyurethane foam, mineral coal (MC), basaltic gravel, and low-density polyethylene. Development of layers in the biofilm carrier surface was evaluated using a flow cell device, and DO profiles were conducted to determine the size of the layers (aerobic and anaerobic). MC was the biofilm carrier selected due to allowing the development of larger aerobic and anaerobic layers in the biofilm (896 and 1,058 μm, respectively). This ability is supposed to improve simultaneous nitrogen removal by nitrification and denitrification biological processes. Thus, as a biofilm carrier, MC was used in a fixed-bed sequencing batch biofilm reactor (FB-SBBR) for treatment of wastewater with a high ammonia concentration (100-400 mgNH-N L). The FB-SBBR (15.0 L) was filled with matrices of the carrier and operated under alternating aeration and non-aeration periods of 6 h each. At a mean nitrogen loading rate of 0.55 ± 0.10 kgNH-N m d, the reactor attained a mean nitrification efficiency of 95 ± 9% with nitrite as the main product (aerobic period). Mean denitrification efficiency during the anoxic period was 72 ± 13%.
A transferência de gases através da interface ar-água é um processo importante para ciclos climáticos de grande escala e para sistemas ambientais menores como rios, lagos, córregos e estações de tratamento de esgoto. Para avançar no entendimento dos princípios básicos envolvidos no fenômeno é necessária a utilização de técnicas e aparatos experimentais adequados. Neste estudo, foram realizadas medidas de concentração através da utilização de micro sonda de oxigênio, em tanque de grade oscilante. A dimensão do elemento sensor da micro sonda é da ordem de alguns micra. Os resultados demonstram a possibilidade de medir, sob condições turbulentas controladas similares às encontradas no ambiente, as flutuações de concentração de oxigênio no interior da camada limite existente imediatamente abaixo da interface ar-água. A quantidade de oxigênio dissolvido nos corpos de água é um indicador primário da qualidade da água. De fato, quando a concentração de oxigênio dissolvido na água cai abaixo de valores aceitáveis, pode afetar significativamente a saúde do ecossistema aquático e também impedir o uso da água para diferentes fins (por exemplo, abastecimento doméstico). A concentração de oxigênio dissolvido na água é o resultado da interação de diversos processos que tendem a aumentar ou diminuir a mesma. A produção fotossintética e a reaeração atmosférica contribuem para o aumento da concentração de oxigênio na água, enquanto que o consumo de oxigênio para a decomposição da matéria orgânica e a respiração realizada pelo ecossistema aquático colaboram para a diminuição da quantidade de oxigênio dissolvido na água. The good composition and activity of biofilms are very important for successful operation and control of fixed-film biological reactors employed in liquid effluents treatment. During the last decade, microsensors have been applied to study microbial ecology. These sensors could provide information regarding the microbial activity concerning nitrification and denitrification that occur inside biofilms. Other techniques of molecular biology, such as fluorescence in situ hybridization (FISH), have also contributed to this matter because their application aids in the identification of the bacterial populations that compose the biofilms. The focus of this paper was to study the loading rate and surface velocity to promote the development of nitrifying biofilms in three distinct flow cells that were employed in the post treatment of a synthetic wastewater simulating the effluent from a UASB (Upflow Anaerobic Sludge Blanket) reactor. Using the FISH technique, it was found that the population of ammonia-oxidizing-bacteria was greater than that of nitrite-oxidizing-bacteria; this was also supported by the lower production of nitrate determined by physicochemical and microsensor analyses. It was verified that the loading rate and surface velocity that promoted the greatest nitrogen removal were 0.25 g N-amon m(-2)biofilm day(-1) and 1 m h(-1), respectively.
