ABSTRAKIsolat golongan senyawa katekin teh hijau (Camelia sinensis) klon GMB4 dapat dikembangkan sebagai agen terapeutik potensial untuk obesitas. Penelitian ini ditujukan untuk mengetahui efek isolat EGCG teh hijau klon GMB4 terhadap ekspresi protein faktor transkripsi C/EBPα dan terhadap kadar protein leptin pada kultur preadiposit viseral tikus. Metode ELISA digunakan untuk mengukur kadar leptin dan immunositokimia untuk ekspresi protein faktor transkripsi C/EBPα. Kultur sel preadiposit viseral tikus yang diisolasi dari tikus Rattus norvegicus Wistar yang ditumbuhkan dalam medium adipogenik dan selanjutnya dipapar dengan isolat golongan senyawa katekin teh hijau (Camelia sinensis) klon GMB4 dengan konsentrasi 0, 50 µM, 100 µM, dan 200 µM. Ekspresi C/EBPα paling rendah terdapat pada konsentrasi isolat EGCG 200 µM. Kadar leptin menurun seiring dengan dosis EGCG yang dinaikkan, sehingga kadar leptin berbanding terbalik dengan dosis EGCG. Kadar leptin terendah pada pemaparan EGCG dengan dosis 200 µM. Dapat disimpulkan bahwa isolat EGCG teh hijau klon GMB4 secara signifikan dapat menurunkan ekspresi protein faktor transkripsi C/EBPα pada kultur preadiposit viseral tikus pada konsentrasi 200 µM dan menurunkan kadar protein leptin pada kultur preadiposit viseral tikus pada konsentrasi 50 µM, 100 µM, dan 200 µM.Kata Kunci: C/EBPα, EGCG, GMB4, leptin, sel preadiposit, teh hijau

Environmental tobacco smoke (ETS) refers to exposure to tobacco smoke, not from our smoking but from being exposed to someone else’s cigarette. Breathing in ETS is known as passive smoking. Cigarette smoke contains dangerous components, include Radical Oxygen Species. Exposure free radical from cigarette result unbalance antioxidant in our body, it can increase protease activity and cell defect. Free radical was inhibited by exogen antioxidants in derivate caffeic acid phenetyl ester (CAPE) propolis Trigona sp. Propolis extract is honey bee product that has many active ingredients with many important roles, as antioxidant, anti-inflammation, DNA protection, and immunomodulator. This research to learn about potential extract propolis Trigona sp for inhibiting increased protease activity and defect of trachea histology in rats exposed to cigarette smoke. It is used five groups, they are negative control, positive control exposed to 2 cigarettes/day, and three preventive groups given by propolis, dose 10, 20 and 30 mg/200gBW/day. We conclude that propolis has the effect to prevent increase protease activity and defect of trachea histology from exposure to cigarette smoke.
The exposure of lead to body can be through inhalation, digestion and adsorption. Lead intoxication can lead to jejuna damages and increase protease activity due to reactive oxygen species (ROS) production. Apple peel extract contains flavonoids that can scavenge ROS. This research aims to investigate the effect of ethanolic extract of Rome Beauty apple peel to protease activity and jejuna histological feature of rat exposed to lead acetate. This research used 20 rats, that divided into 5 treatment groups, which were negative control group (healthy rat), positive control group (exposed to lead acetate 10 mg/rat/day for 14 days), and preventive therapy group (administrated the apple peels extract with doses of 28 mg/200 g BW, 56 mg/200g BW, 112mg/200g BW for 21 days and also lead acetate with 10mg/rat/day for 14 days on day 15th until 28th). Jejunal histological features was stained using HE stain which then observed using a microscope. Protease activity was measured by using spectrophotometric method. The data of protease activity was statistically analyzed using ANOVA, followed by Tukey test (α = 5%). The results showed that the administration of ethanolic extract of Rome Beauty apple peels at best dose of 112 mg/200 g BW can reduce protease activity and improve jejuna histopathology. The conclusion of this study was that apple peels extract of Rome Beauty can be used to prevent the increase of protease activity and the damage of rat jejuna exposed to lead acetate.
