Oil cakes have excellent nutritional value and offer considerable potential for use in biotechnological processes that employ solid-state fermentation (SSF) for the production of high value products. This work evaluates the feasibility of using canola cake as a substrate for protease production by a selected strain of Aspergillus oryzae cultivated under SSF. The influences of the following process parameters were considered: initial substrate moisture content, incubation temperature, inoculum size, and pH of the buffer used for protease extraction and activity analysis. Maximum protease activity was obtained after cultivating Aspergillus oryzae CCBP 001 at 20°C, using an inoculum size of 107 spores/g in canola cake medium moistened with 40 mL of water to 100 g of cake. Cultivation and extraction under selected conditions increased protease activity 5.8-fold, compared to the initial conditions. Zymogram analysis of the enzymatic extract showed that the protease molecular weights varied between 31 and 200 kDa. The concentrated protease extract induced clotting of casein in 5 min. The results demonstrate the potential application of canola cake for protease production under SSF and contribute to the technological advances needed to increase the efficiency of processes designed to add value to agroindustrial wastes.
In this study, we investigated the main constituent, the predominant class and biological activity of the essential oil extracted from the leaves of Pimenta dioica and the pattern of the major constituent against larvae in the third stage of Aedes aegypti. For this reason, we extracted the oil by hydrodistillation, identified its components by gas chromatography coupled with mass spectrometry (GC/MS) and calculated the lethal concentration (LC50) of the larvicidal activity using the Reed-Muench method. The results show that the oil consists mainly of eugenol, in which the phenylpropanoid class predominated and the lethal concentration, LC50, was 38.86 μg mL-1at a confidence level of 2.25 μg mL-1, while the eugenol standard presented LC5079.75 μg mL-1at a confidence level of 2.10 μg mL-1. Given the facts, we conclude that the oil is more active than the standard and that it has the potential to replace chemical larvicides.
O estudo teve como objetivo verificar o nível de conhecimento dos alunos do ensino médio de uma escola da rede pública acerca de primeiros socorros e suas reações mediante as necessidades de urgência e emergência. Método: trata-se de um estudo transversal de natureza aplicada, com caráter quantitativo. Quanto aos fins da pesquisa caracteriza-se como descritiva, sendo a obtenção de dados realizada através da aplicação de um formulário aos estudantes do 1º ao 3º ano do Ensino Médio. Resultados: Participaram do estudo 167 alunos, de ambos os sexos, com idade variável entre 15 e 18 anos, sendo que desses 95% relataram que o ambiente escolar é sim propício a ocorrência de acidentes e que possam necessitar de assistência de primeiros socorros imediata, e ainda segundo esses 37,7% já passaram por situações em que era necessária intervenção rápida, mas que nessas situações 50,2% não sabia o que fazer. Conclusão: Foi possível perceber a importância da capacitação desses alunos a respeito de primeiros socorros, tendo em vista o ambiente o qual os mesmos estão expostos todos os dias e como esses conhecimentos podem ser eficazes e crucias diante de situações de pessoas vítimas de circunstância emergenciais.
