Human immunodeficiency virus (HIV) infections continue to exert an enormous impact on global human health. This led experts to emphasize the importance of new measures for preventing HIV infections, including the development of vaccines and novel drugs. In this context, a promising approach involves the use of lectins that can bind the surface envelope glycoprotein gp120 of HIV with high affinity, preventing viral entry. The cyanobacterial lectin microvirin (MVN) has been proposed as a candidate for development as a topical microbicide because of its ability to bind to high mannose-type glycans, potently inhibiting HIV-1 entry. Thus, the aim of this computational study was to investigate the effects of four point mutations (D53Q, D53E, D53K, and D53W) on the structure and affinity of MVN with di-mannose (MAN). Molecular dynamics simulations followed by binding free energy calculations using MM-GBSA were employed. The calculated binding free energy of ligand-receptor complexation of MVN with MAN was -26.02 kcal mol. We identified in the wild-type protein that residues I45, T59, and Q81 have a major contribution to the binding free energy of di-mannose. Among the investigated mutants, the most promising one was the D53W mutation, with a theoretical binding free energy value of -29.16 kcal mol. We suggest that this increased stability is due to the introduction of extra rigidity on the hinge region connecting two key structural elements of the MVN binding site.
Asparaginases are found in a range of organisms, although those found in cyanobacteria have been little studied, in spite of their great potential for biotechnological application. This study therefore sought to characterize the molecular structure of an L-asparaginase from the cyanobacterium Limnothrix sp. CACIAM 69d, which was isolated from a freshwater Amazonian environment. After homology modeling, model validation was performed using a Ramachandran plot, VERIFY3D, and the RMSD. We also performed molecular docking and dynamics simulations based on binding free-energy analysis. Structural alignment revealed homology with the isoaspartyl peptidase/asparaginase (EcAIII) from Escherichia coli. When compared to the template, our model showed full conservation of the catalytic site. In silico simulations confirmed the interaction of cyanobacterial isoaspartyl peptidase/asparaginase with its substrate, β-Asp-Leu dipeptide. We also observed that the residues Thr154, Thr187, Gly207, Asp218, and Gly237 were fundamental to protein-ligand complexation. Overall, our results suggest that L-asparaginase from Limnothrix sp. CACIAM 669d has similar properties to E. coli EcAIII asparaginase. Our study opens up new perspectives for the biotechnological exploitation of cyanobacterial asparaginases.
Introdução: Asparaginases são amino-hidrolases que catalisam a hidrólise de asparagina em aspartato e amônia. Desde 1953, estas enzimas são conhecidas por sua atividade anticancerígena devida à dependência, que alguns tecidos tumorais têm de Lasparagina extracelular para sua proliferação. Assim, uma vez injetadas na corrente sanguínea, as L-ASNases reduzem a quantidade de asparagina no corpo, impedindo a sobrevivência das células tumorais. Objetivo: Caracterizar o potencial biotecnológico das L-asparaginases de Escherichia coli por meio de ferramentas de simulação computacional. Material e métodos: A enzima Asparaginase tipo II (ECAII) foi obtida no Protein Data Bank - PDB. Dos PDBs 1IHD, 1JAZ, 1JJA, 1NNS, 1HO3, 4ECA e 3ECA, foi escolhida a enzima tipo selvagem mais antiga depositada sob o código 3ECA. Todas as demais são formas mutantes da molécula. Para o estudo e visualização do sítio da enzima foram utilizados os softwares PYMOLL e Visual Molecular Dynamics em suas versões mais recentes. Resultados e discussão: A estrutura de ativa de ECAII é um tetrâmero com 4 subunidades: A, B, C e D, cada um com um sítio ativo. Consiste de dois domínios α e β, Nt-terminal e C-terminal. O domínio C-terminal vai até o aminoácido Gln190, seguido por alça que conecta a outra extremidade, que vai do resíduo 191 ao 212, seguida da extremidade C-terminal, mais curta, com apenas 113 resíduos. Foram identificadas as cavidades do sítio da enzima e quais resíduos interagem com o substrato asparagina. Conclusão: As caractetísticas moleculares e interação com o substrato observada no sítio ativo da molécula confirmam o potencial biotecnológico dessas enzimas. Tendo isso em vista, sugere-se que as asparaginases sejam otimizadas por meio de mutações em seu sítio ativo, aumentando sua atividade catalítica e, consequentemente, sua atividade anticancerígena.
A microbiota vaginal possui papel importante no equilíbrio, na manutenção e prevenção da saúde vaginal das mulheres, possuindo vasta quantidade de microrganismos. Candida spp. é uma das espécies de fungos que mais causa infecções nos seres humanos. Durante a gravidez, onde os níveis hormonais estão elevados e ocorre variação do pH vaginal, esses microrganismos podem colonizar a vagina e consequentemente propiciar uma infecção. O presente trabalho visa avaliar os aspectos da microbiota vaginal e sua correlação com a candidíase vulvovaginal na gravidez, e suas formas de diagnóstico. O presente estudo caracteriza-se por ser uma revisão integrativa de literatura, onde foram utilizadas as plataformas científicas PubMed, Google Acadêmico, Medline e Scielo Artigos como fonte de dados. Os dados gerados na pesquisa demonstraram a importância da compreensão sobre a microbiota vaginal. Além de evidenciar a importância de estudos a respeito da candidíase vulvovaginal em gestantes, e como outras espécies de Candida sp. estão afetando e elevando os índices de pacientes com corrimento vaginal, além da Candida albicans, a qual é a espécie mais estudada desse gênero. Também observar a relevância do diagnóstico dessa enfermidade. Concluímos assim que o conhecimento dos aspectos clínicos da sintomalogia, principalmente, durante a gravidez é uma importante ferramenta para o diagnóstico precoce durante o período pré-natal. A transmissão vertical se mostra presente nas infecções em recém-nascido. A espécie mais evidente desse gênero é a C. albicans contudo, as espécies de Candida não albicans estão cada vez mais presentes nas infecções causadas pelo gênero Candida spp.
