Scopulariopsisand scopulariopsis-like species from indoor environments

Abstract: Scopulariopsis-like species are often reported from the indoor environment, as well as from clinical samples. The lack of type isolates and thorough phylogenetic studies in the Microascaceae hampered the correct identification of these isolates. Based on recent phylogenetic studies, which resulted in multiple name changes, the aim is to molecularly identify the Scopulariopsis and scopulariopsis-like species which occur in the indoor environment and give an overview of the current species in these genera and th… Show more

“…DNA extractions were performed using the Ultraclean ® Microbial DNA Isolation Kit (MoBio laboratories, Carlsbad, CA, USA), following manufacturer's instructions. The internal transcribed spacer 1 and 2 and intervening 5.8S nrDNA (ITS), and parts of the β-tubulin ( tub2 ) and translation elongation factor 1-α ( tef1 ) genes were amplified and sequenced as described in Woudenberg et al. (2017) .…”

“…The best nucleotide substitution models were determined with Findmodel ( http://www.hiv.lanl.gov/content/sequence/findmodel/findmodel.html ). For both the single gene sequence alignments and the concatenated alignment, Bayesian and Maximum-likelihood analyses were performed as described in Woudenberg et al. (2017) .…”

“…Microascus spp. are ubiquitous environmental saprotrophic ascomycetes that are usually isolated from soil or decaying plant material, but can also occur in indoor environments . Traditionally, their asexual morphs were classified as Scopulariopsis .…”

“…Phylogenetic reconstructions by maximum likelihood (ML) and bayesian inference (BI) approaches were performed with MEGA v. 6.0 and MrBayes v. 3.2, respectively. The strain Pithoascus stoveri CBS 176.71 was selected as outgroup . The combined analysis of the four phylogenetic markers was tested through Incongruence Length Difference (ILD) implemented in Winclada programme …”

“…Of these, about 90% are associated with one of the following risk factors: AIDS, organ transplantation, corticosteroid therapy, peritoneal dialysis, surgery, cardiac disease, and trauma. Scopulariopsis is a large genus of saprophytic organisms found mostly in soil, but frequently isolated from food, paper, and other materials [3]. Within the genus are members of both hyalohyphomycoses and phaeohyphomycoses.…”
